丙戊酸处理对猪体细胞克隆胚胎体外、体内发育效率的影响

摘要 为了探讨丙戊酸(valproic acid, VPA)处理对猪体细胞克隆胚胎体外、体内发育效率的影响,提高猪体细胞克隆效率,试验比较了VPA不同处理浓度(2,4,6 mmol/L)和时间(18,24,48 h)对克隆胚胎体外发育的影响,并将VPA处理的克隆胚胎移植给代孕母猪,比较对照组和VPA处理组的妊娠率、分娩率、窝均总仔数、克隆效率、平均单头代孕母猪产活仔数,验证VPA处理对克隆胚胎体内发育效率的影响。结果表明:用2 mmol/L VPA处理克隆胚胎24 h可显著提高囊胚率(P<0.05),但对卵裂率和囊胚内细胞数无显著影响(P>0.05)。VPA处理组代孕母猪妊娠率高于对照组,分娩率、窝均总仔数、窝均活仔数、克隆效率、平均单头代孕母猪产活仔数均低于对照组,但差异不显著(P>0.05)。说明用2 mmol/L VPA处理克隆胚胎24 h可显著提高囊胚率,对妊娠率、分娩率、窝均总仔数、窝均活仔数、克隆效率、平均单头代孕母猪产活仔数无影响。 To examine the effects of valproic acid(VPA) treatment on capacity of porcine cloned embryos in vitro and in vivo, and improve the efficiency of pig somatic cell cloning, the effects of VPA different concentration(2,4,6 mmol/L) and different processing time(18,24,48 h) on in vitro culture of pig cloned embryos were compared;and cloned embryos treated with VPA were transferred to surrogate sows to compare the pregnancy rate, delivery rate, total litter size, cloning efficiency, and average live litter size of single surrogate sows in the control group and the treatment group, so as to verify the influence of VPA treatment on the development efficiency of cloned embryos in vivo. The results showed that the blastocyst rate of pig cloned embryos treated with 2 mmol/L VPA at 24 h was significantly increased(P<0.05), but 2 mmol/L VPA had no significant effect on the cleavage rate and the total number of cells in the blastocyst(P>0.05). Pregnancy rate of surrogate sows in the experimental group and was higher than that in the control group. There were not significant differences in pregnancy rate, delivery rate, total litter size per litter, live litter size per litter, cloning efficiency and average live litter size per sow between the treatment group and the control group(P>0.05). It indicated that the cloned embryos of pigs treated with 2 mmol/L VPA for 24 h could significantly improve the blastocyst rate, but it had no effect on pregnancy rate, delivery rate, total litter size per litter, live litter size per litter, cloning efficiency, and average live litter size per single surrogate sow.
