摘要
目的探讨大豆苷对A549细胞增殖和迁移的调节作用。方法将DMSO、50、100和150μmol·L-1大豆苷作用于A549细胞,通过CCK8观察大豆苷对细胞增殖的影响,将浓度为0~200μmol·L-1的大豆苷作用于A549细胞,计算大豆苷对于A549细胞的IC50,采用流式细胞技术分析大豆苷对细胞周期的影响;采用transwell实验检测浓度为50、100和150μmol·L-1的大豆苷作用24 h后对A549细胞迁移能力的影响;采用Western blot实验检测大豆苷作用于A549细胞后STAT3p-Tyr705、STAT3、Cyclin D1和MMP2蛋白的表达情况。结果CCK8检测发现50、100和150μmol·L-1大豆苷作用于A549细胞后均能显著抑制细胞的增殖情况,大豆苷对于A549细胞作用24 h的IC50约为148.8μmol·L-1,大豆苷能够抑制细胞周期G1/S期的进程。Transwell实验结果显示大豆苷能够显著抑制A549细胞的转移功能。Western blot实验结果显示大豆苷抑制A549细胞中STAT3p-Tyr705、Cyclin D1和MMP2的表达但不影响STAT3的表达。结论大豆苷能够通过抑制STAT3的磷酸化水平抑制肺癌细胞系A549细胞的增殖和迁移。
Objective To investigate the effects of Daidzein on proliferation and migration of A549 cells.Methods A549 cells were treated with DMSO,50,100 and 150 μmol·L-1 daidzein.The effect of Daidzein on cell proliferation was observed by CCK8.IC50 of A549 cells was calculated by 0-200μmol·L-1 daidzein.The effect of Daidzein on cell cycle was analyzed by flow cytometry.The migration ability of A549 cells was detected by Transwell after 24 h of 50,100 and 150 μmol·L-1 daidzein.Western blot assay was used to detect the expression of STAT3p-Tyr705,STAT3,Cyclin D1 and MMP2 proteins in A549 cells treated with daidzein.Results CCK8 assay showed that 50,100 and 150 μmol·L-1 daidzein could significantly inhibit the proliferation of A549 cells.The IC50 of Daidzein on A549 cells for 24 h was about 148.8 μμmol·L-1.Daidzein could inhibit the process of G1/S phase of cell cycle.Transwell assay showed that daidzein could significantly inhibit the metastasis of A549 cells.Western blot results showed that daidzein inhibited the expression of STAT3p-Tyr705,Cyclin D1 and MMP2 in A549 cells,but did not affect the expression of STAT3.Conclusion Daidzein can inhibit the proliferation and migration of lung cancer cell line A549 by inhibiting the phosphorylation of STAT3.
