相分离理论在栀子豉汤提取物制备中的应用

Application of phase separation theory in the extraction preparation of zhizi-chi decoction

目的应用相分离理论对含果实类中药的方剂进行提取、纯化、浓缩、干燥等过程的研究,通过药效物质在固-液两相中的传递,达到提高提取率,并有效除去油脂、蛋白质等杂质,在此基础上浓缩、干燥制备提取物,达到去粗存精的目的。方法以栀子豉汤为模型药,以干燥提取物中栀子苷、以染料木素计总黄酮的含量及出膏量为测定指标进行考察,在多种提取条件下分别试验,对试验结果用相分离理论加以分析。结果取处方量药材,加入体积分数60%乙醇溶液浸泡2h,在60℃下动态低温提取0.5 h,滤过,采用壳聚糖聚合沉降法去除蛋白质,离心法去除油脂等杂质,减压浓缩并干燥,得提取物1.094 5 g,其中栀子苷含量为45.56 mg·g,总黄酮含量为8.98 mg·g。结论相分离理论可有效指导含果实类中药方剂提取物制备过程,达到去粗存精的目的。

Objective Applying phase separation theories to study the processes of extraction,purification concentration and drying with seed-fruit Chinese medicine.Through the transfer of medicinal materials in solid-liquid phase,the extraction rate can be improved,and impurities such as oil and protein can be effectively removed.On this basis,the extract can be concentrated and dried to achieve the purpose of discarding the dross and keeping the finer part.Methods zhizi-chi decoction was used as a model drug,the content of geniposide and total flavone calculated by genistein and amount of cream in dry extract were used as the determination indexes.Experiment was tested under several extract conditions.The result of experiment was analyzed by phase separation theory.Results prescription dosage of medicinal material soaked in 60% alcohol solution for 2 h,and for 0.5 h in dynamic immersion under 60 ℃.when the filtrate had been centrifuged to remove part the grease,and remove protein and other impurities by chitosan polymerization sedimentation method,the extract was reduced pressure,concentrated and dried to obtain 1.0945 g.the geniposide was 45.5601 mg·g-1 the total flavonoid content was 8.9825 mg·g-1.Conclusion phase separation theory can guide the extraction preparation process of seed-fruit Chinese medicinal formula to achieve the purpose of discarding the dross and keeping the finer part.
