广西地区犬、猫细环病毒的全基因组序列分析

Whole genome sequence analysis of Torque teno canis virus and Torque teno felis virus in Guangxi region

摘要 为了解细环病毒(Torque teno virus, TTV)在广西地区犬、猫中的流行情况,试验利用PCR方法对采集自广西地区的58份犬、猫腹泻样品进行检测,并对阳性样品进行全基因组序列扩增、测定及遗传进化分析。结果表明:犬细环病毒(TTCaV)阳性率为8.57%(3/35),猫细环病毒(TTFeV)阳性率为8.70%(2/23)。成功获得2株TTCaV(GXNN32和GXNN34)和2株TTFeV(GX26和GX27)的全长基因组序列。2株TTCaV全基因组序列与TTCaV参考株cf-TTV10(日本)、GX206(中国)同源性为96.2%~98.5%,与猪源、人和非人灵长类TTV同源性为18.0%~50.8%;2株TTFeV的全基因组序列与TTFeV参考毒株Fc-TTV4(日本)、PRA4(法国)、SH-F1(中国)、TTFeV278(捷克共和国)同源性为92.3%~96.7%,与猪、人和非人灵长类TTV同源性18.3%~52.8%。本研究获得的2株TTCaV和2株TTFeV之间的同源性为33.9%~34.0%。说明本研究获得的TTCaV和TTFeV分别属于辛型细环病毒属(Thetatorquevirus)和庚型细环病毒属(Etatorquevirus),2株TTCaV和2株TTFeV之间的同源性为33.9%~34.0%。犬、猫可感染特定物种TTV,并且基因组大小存在差异。 In order to understand the prevalence of Torque teno virus(TTV) in dogs and cats in Guangxi Zhuang Autonomous Region, PCR method was used in this experiment to analyze 58 diarrhea samples of dogs and cats collected from Guangxi Zhuang Autonomous Region, and the positive samples were subjected to whole genome sequence amplification, determination and genetic evolution analysis. The results showed that the positive rate of Torque teno canis virus(TTCaV) was 8.57%(3/35),and the positive rate of Torque teno felis virus(TTFeV) was 8.70%(2/23). The full-length genome sequences of two strains of TTCaV(GXNN32 and GXNN34) and two strains of TTFeV(GX26 and GX27) were successfully obtained. The analysis found that the two strains of TTCaV whole genome sequences had 96.2%-98.5% homology with the TTCaV reference strains cf-TTV10(Japan) and GX206(China),and had 18.0%-50.8% homology with pig, human and non-human primate-derived TTV. The whole genome sequences of the two TTFeV strains had 92.3%-96.7% homology with the reference strains of TTFeV Fc-TTV4(Japan),PRA4(France),SH-F1(China),and TTFeV278(Czech Republic),and had 18.3%-52.8% homology with pig, human and non-human primate-derived TTV. The homology between the two TTCaV strains and the two TTFeV strains obtained in this study was 33.9%-34.0%.The results suggested that the TTCaV and TTFeV belong to the genus Thetatorquevirus and Etatorquevirus, respectively. The homology between the 2 strains of TTCaV and the 2 strains of TTFeV is 33.9%~34.0%. Dogs and cats can be infected with specific TTV species, and TTV genome sizes were different.
