摘要
目的探讨壳寡糖对子宫内膜癌细胞增殖迁移的影响及可能分子机制。方法将不同浓度的壳寡糖作用于子宫内膜癌HEC-1B细胞,通过CCK-8检测细胞的增殖情况;划痕实验检测细胞迁移情况;Western blot检测E-cadherin、Snail、MMP2和MMP9及AMPK/mTOR的蛋白表达。结果 1.25mg/ml的壳寡糖对HEC-1B细胞已有增殖抑制作用,且呈浓度依赖性与时间依赖性;壳寡糖抑制HEC-1B细胞迁移;壳寡糖能上调E-cadherin的蛋白表达,抑制Snail、MMP2和MMP9的表达;激活AMPK信号,抑制mTOR的蛋白表达。结论壳寡糖抑制子宫内膜癌细胞的增殖和迁移,可能与壳寡糖激活AMPK信号并抑制mTOR蛋白以及E-cadherin表达相关。
Objective To investigate the effect of chitosan oligosaccharide on the proliferation and migration of endometrial cancer cells and its possible molecular mechanism. Methods Different concentrations of chitosan oligosaccharides were treated on HEC-1B cells. The proliferation of HEC-1B cells was detected by CCK-8. The migration of HEC-1B cells was detected by scratch test. The protein expression of E-cadherin, snail, MMP2, MMP9 and AMPK/mTOR was detected by Western blot. Results 1.25 mg/ml chitosan inhibited the proliferation of HEC-1 B cells, and it was concentration dependent and time-dependent. Chitosan inhibited the migration of HEC-1B cells. Chitosan could up regulate the protein expression of E-cadherin, inhibit the expression of snail, MMP2 and MMP9;activate AMPK signal and inhibit the protein expression of mTOR. Conclusion Chitosan inhibit the proliferation and migration of endometrial cancer cells, which may be related to the activation of AMPK signal and inhibition of mTOR protein and E-cadherin expression.
