摘要
目的研究人肝癌细胞97H中Yes相关蛋白(Yes-Associated Protein,YAP)mRNA的下调对TGF-β1 mRNA的影响。方法首先以RNA干扰技术下调人肝癌细胞97H的YAP mRNA水平,用PCR技术检测转化生长因子-β1(transforming growth factor-β1,TGF-β1)mRNA的水平变化。其次,用PCR技术检测人肝癌细胞97H与97L细胞中TGF-β1 mRNA水平的差别。结果人肝癌细胞97H细胞经RNA干扰使YAP mRNA水平下调后,细胞内的TGF-β1 mRNA水平升高。自然存在的表达YAP较低的97L细胞中TGF-β1 mRNA水平较97H高。结论人肝癌细胞97H中YAP下调过程伴随着TGF-β1 mRNA同步升高,对该细胞YAP基因的抑制可导致内源性TGF-β1升高。
Objective To study the effect of down-regulation of YAPmRNA on TGF-β1 mRNA in human hepatocellular cancer cell line 97H.Methods RNA interference technology was used to downregulate YAPmRNA levels in MHCC97H cells.Changes of TGF-β1 mRNA expression levels were detected by PCR.PCR was used to detect the difference in TGF-β1 mRNA levels between MHCC97H and MHCC97L cells.Results After the YAPmRNA level of MHCC97H cells was down-regulated by RNA interference technology,the level of TGF-β1 mRNA in the cells increased.Naturally-expressed MHCC97L cells with lower YAP expression had higher TGF-β1 mRNA levels than MHCC97H.Conclusion The down-regulation of YAP in human hepatoma cells MHCC97H is accompanied by a simultaneous increase in TGF-β1 mRNA.Inhibition of the YAP gene in these cells can lead to an increase in endogenous TGF-β1.
