Effect of Nerve Growth Factor on the Development and Poor Prognosis of Colorectal Cancer

目的探讨神经生长因子(NGF)表达与结直肠癌临床病理特征和生存率的关系。方法选取2018年1—6月益阳市人民医院石蜡包埋的结直肠癌样本96例,2020年6-7月成都医学院第二附属医院新鲜结直肠癌组织及癌旁正常组织样本5例。采用蛋白质印迹法和免疫组织化学方法,检测结直肠癌组织中NGF的表达情况,分析NGF表达量与结直肠癌病理特征的关系,利用癌症基因图谱(TCGA)数据库分析NGF表达情况与结直肠癌患者生存率的关系。结果NGF在结直肠癌组织中表达上调,淋巴结无转移和TNM分期(Ⅰ~Ⅱ)患者NGF阳性表达率低于淋巴结转移和TNM分期(Ⅲ~Ⅳ)(P<0.001),且NGF高表达的生存率低于NGF低中表达的生存率(P=0.037)。结论NGF促进了结直肠癌的发展和不良预后。

Objective To investigate the relationship between the expression of nerve growth factor(NGF)and clinicopathological characteristics,as well as survival rate of colorectal cancer.Methods Ninety-six paraffinembedded colorectal cancer samples from Yiyang People’s Hospital from January to June 2018 were selected.And 5 fresh colorectal cancer tissue samples and adjacent normal tissue samples were selected from the Second Affiliated Hospital of Chengdu Medical College from June to July 2020.Western blotting and immunohistochemistry were used to detect the expression of NGF in colorectal cancer tissues.The correlation between the expression of NGF and the pathological characteristics of colorectal cancer was statistically analyzed.And the relationship between NGF expression and survival rate of colorectal cancer patients was mined by the Cancer Genome Atlas(TCGA)database.Results The expression of NGF was up-regulated in colorectal cancer tissues.The positive expression rate of NGF in patients without lymph node metastasis and tumor node metastasis(TNM)staging(Ⅰ~Ⅱ)was lower than that in patients with lymph node metastasis and TNM staging(Ⅲ~Ⅳ)(P<0.001).And the expression of NGF was negatively correlated with survival rate(P=0.037).Conclusion NGF promotes the development and poor prognosis of colorectal cancer.
