摘要
目的研究玉屏风颗粒对组胺(histamine)和聚肌胞苷酸[Poly(I:C)]分别诱导HaCaT特应性皮炎细胞模型的作用及其相关细胞炎症因子表达的影响。方法以不同浓度的玉屏风颗粒处理正常HaCaT细胞,MTT法检测该药对细胞存活率的影响;使用histamine和Poly(I:C)分别处理HaCaT细胞,构建不同诱导剂引起的特应性皮炎细胞模型,MTT法检测不同浓度的玉屏风颗粒对造模后HaCaT细胞存活率的影响,实时荧光定量PCR(RT-qPCR)检测玉屏风颗粒对造模后HaCaT细胞中相关炎症因子mRNA的表达。结果与空白组比较,50、25、12.5μg/L质量浓度的玉屏风颗粒能显著提高正常HaCaT细胞的存活率(P<0.01或P<0.05);HaCaT细胞经histamine诱导之后,与模型组比较,50、25、12.5、6.25μg/L质量浓度的玉屏风颗粒能显著提高细胞的存活率(P<0.01);50μg/L的玉屏风颗粒显著抑制IL-1β、IL-6、IL-8、TNF-α和MCP-1的mRNA表达(P<0.01或P<0.05),25μg/L的玉屏风颗粒显著抑制IL-1β和TNF-α的表达(P<0.01或P<0.05);HaCaT细胞经Poly(I:C)诱导之后,与模型组比较,50、25μg/L的玉屏风颗粒能显著提高细胞的存活率(P<0.01);50μg/L的玉屏风颗粒能显著抑制IL-1β、IL-8、TNF-α和MCP-1的mRNA表达(P<0.01或P<0.05),25μg/L的玉屏风颗粒显著抑制IL-1β、IL-6、IL-8、TNF-α的mRNA表达(P<0.05),12.5μg/L的玉屏风颗粒显著抑制MCP-1的mRNA表达水平(P<0.01)。结论玉屏风颗粒可改善HaCaT特应性皮炎细胞模型的细胞增殖并抑制相关细胞炎症因子的表达。
Objective To study the effect of Yupingfeng granule on HaCaT atopic dermatitis cell model induced by histamine and poly(I:C) and the expression of related inflammatory cytokines.Methods Normal HaCaT cells were treated with different concentrations of Yupingfeng granules.The effect of Yupingfeng granules on cell survival rate was detected by MTT assay.HaCaT cells were treated with histamine and poly(I:C) respectively to establish cell models of atopic dermatitis.MTT method was used to detect the effect of different concentrations of Yupingfeng granules on the survival rate of HaCaT cells after modeling,and real-time quantitative PCR was used to detect the mRNA expression of related inflammatory factors in HaCaT cells after modeling.Results Compared with the blank group,Yupingfeng granules at the concentrations of 50,25 and 12.5 μg/L significantly increased the survival rate of normal HaCaT cells(P<0.01 or P<0.05).Compared with the model group,Yupingfeng granules at the concentrations of 50,25,12.5 and 6.25 μg/L significantly improved the survival rate of HaCaT cells induced by histamine(P<0.01),Yupingfeng granules at a concentration of 50 μg/L significantly inhibited the mRNA expression of IL-1β,IL-6,IL-8,TNF-α and MCP-1(P<0.01 or P<0.05),and Yupingfeng granules at a concentration of 25 μg/L significantly reduced the expression of IL-1β and TNF-α(P<0.01 or P<0.05).Compared with the model group,Yupingfeng granules at the concentrations of 50 and 25 μg/L significantly improved the survival rate of HaCaT cells induced by Poly(I:C)(P<0.01),Yupingfeng granules at a concentration of 50 μg/L significantly inhibited the mRNA expression of IL-1β,IL-8,TNF-α and MCP-1(P<0.01 or P<0.05),Yupingfeng granules at a concentration of 25 μg/L significantly reduced the expression of IL-1β,IL-6,IL-8,and TNF-α(P<0.05),and Yupingfeng granules at a concentration of 12.5 μg/L significantly decreased MCP-1 mRNA expression(P<0.01).Conclusion Yupingfeng granules can improve the cell proliferation and inhibit the expression of inflammatory factors in HaCaT cell model of atopic dermatitis.
