摘要
目的探讨DAPK3和CDK8在宫颈癌组织的表达情况,为宫颈癌的诊断和预后指示提供辅助依据。方法选取行手术治疗的68例宫颈癌患者为研究对象,收集宫颈癌组织和癌旁正常组织,进行免疫组织化学法检测。分析DAPK3和CDK8在宫颈组织中的表达情况、与不同临床特征的关系、与宫颈癌患者预后的关系。结果宫颈癌组织中DAPK3低表达率为98.53%,显著高于癌旁正常组织的8.82%,CDK8在宫颈癌组织中的阳性表达率为92.65%,显著高于癌旁正常组织的10.29%(P>0.05);不同年龄段、不同FIGO分期、不同组织学类型和分化程度的DAPK3和CDK8的表达情况无统计学差异(P>0.05);单因素回归分析显示,DAPK3(P=0.031)低表达较之高表达,CDK8(P=0.040)高表达较之低表达,显著增加宫颈癌患者的死亡风险,ⅠB2~ⅡA期(P=0.000)和低分化程度(P=0.028)也是患者死亡的风险因素。多因素回归分析显示,DAPK3低表达(P=0.004)、CDK8高表达(P=0.017),与患者的不良预后密切相关,可作为独立预测指标。结论宫颈癌患者宫颈癌组织中DAPK3呈低表达状态,CDK8呈高表达状态,低水平的DAPK3和高水平的CDK8提示患者预后不良。
Objective To explore the expression of DAPK3 and CDK8 in cervical cancer tissues,and to provide an auxiliary basis for the diagnosis and prognostic indication of cervical cancer.Methods 68 cases of cervical cancer patients undergoing surgical treatment were selected as the research objects.Cervical cancer tissue and normal tissues adjacent to the cancer were collected for immunohistochemical detection.Analyze the expression of DAPK3 and CDK8 in cervical tissue,the relationship with different clinical features,and the relationship with the prognosis of cervical cancer patients.Results The low expression rate of DAPK3 in cervical cancer tissues was 98.53%,which was significantly higher than 8.82%in normal tissues adjacent to cancer,and the positive expression rate of CDK8 in cervical cancer tissues was 92.65%,which was significantly higher than 10.29%in normal tissues adjacent to cancer(P>0.05);There was no statistically significant difference in the expression of DAPK3 and CDK8 in different age groups,different FIGO stages,different histological types and degrees of differentiation(P>0.05);Single factor regression analysis showed that the low expression of DAPK3(P=0.031)was higher than that of CDK8(P=0.040),which significantly increased the risk of death of cervical cancer patients.StageⅠB2~ⅡA(P=0.000)and poor differentiation(P=0.028)were also risk factors for patients’death.Multivariate regression analysis showed that low expression of DAPK3(P=0.004)and high expression of CDK8(P=0.017)were closely related to the poor prognosis of patients and can be used as independent predictors.Conclusion Cervical cancer patients have low expression of DAPK3 and high expression of CDK8 in cervical cancer tissues.Low levels of DAPK3 and high levels of CDK8 indicate a poor prognosis.
