补阳还五汤对气虚血瘀模型大鼠血液流变学的影响 被引量:6

摘要 目的:研究补阳还五汤对气虚血瘀模型大鼠血液流变学相关指标变化的影响。方法:选用SD清洁级大鼠40只,雌雄各半,随机分为空白组、模型组以及补阳还五汤高(14.0 g/kg)、低(3.5 g/kg)剂量组,每组10只大鼠。除空白组外,其他各组大鼠均气管内一次性注射0.9%氯化钠溶液配制的博莱霉素复制气虚血瘀动物模型(7 mg/kg)。造模14 d后,补阳还五汤高、低剂量组连续灌胃给药14 d,末次给药1 h后5%水合氯醛麻醉,腹主动脉取血,对全血黏度、血浆黏度、凝血四项、红细胞压积、聚集、刚性、变形等相关指标进行检测。结果:与空白组比较,模型组大鼠各切变率全血黏度显著升高(P<0.05),红细胞压积、红细胞聚集指数及红细胞刚性指数均明显增加(P<0.05),红细胞变形指数显著降低(P<0.05),APTT、TT、PT时间延长,FIB含量明显增加;与模型组比较,补阳还五汤的高、低剂量组均能明显降低高、中、低切的全血黏度(P<0.05),显著降低血浆黏度(P<0.05),减轻红细胞压积、降低FIB含量、抑制红细胞聚集能力、增强红细胞变形能力。结论:补阳还五汤可以显著降低气虚血瘀模型大鼠的血液黏度,改善血液浓、黏、凝、聚状态。 Objective:To study the effect of Buyang Huanwu Decoction on the changes of hemorheology-related indexes in rats with Qi deficiency and blood stasis.Methods:40 SD rats were randomly divided into the blank group,the model group and the Buyang Huanwu Decoction groups of high-dose(14.0 g/kg)and low-dose(3.5 g/kg),with 10 rats in each group.The animal model of Qi deficiency and blood stasis was established by intratracheally injecting with Bleomycin prepared by 0.9%Sodium Chloride solution(7 mg/kg).After 14 days of modeling,Buyang Huanwu Decoction of high-dose and low-dose were continuously intragastricaly administrated for 14 days,one hour after the last administration,5%Chloral Hydrate was anesthetized,and blood was taken from the abdominal aorta for the detection of whole blood viscosity,plasma viscosity,four coagulation indicators,hematocrit,aggregation,rigidity,deformation and other related indicators.Results:The whole blood viscosity of each shear rate was significantly increased(P<0.05),hematocrit,aggregation indexes and rigidity indexes were obviously increased(P<0.05),and the deformation indexes were significantly decreased(P<0.05),APTT,TT and PT were prolonged,FIB content was remarkably increased in the model group compared to those in the blank group.Compared to those in the model group,the high-shear,medium-shear and low-shear of whole blood viscosity were significantly decreased(P<0.05),plasma viscosity was also obviously reduced(P<0.05),hematocrit and FBI content were decreased,red blood cell aggregation was inhibited,and red blood cell deformability was enhanced in the Buyang Huanwu Decoction groups of high-dose and low-dose.Conclusion:Buyang Huanwu Decoction can significantly reduce the blood viscosity of rats with Qi deficiency and blood stasis,and improve the state of blood concentration,viscosity,coagulation and aggregation.
