lncRNA FENDRR Inhibiting the Proliferation,Migration and Invasion of Glioblastoma Cells by Targeting MiR-362-5p

摘要 目的:探讨FOXF1毗连非编码发育调控RNA(FENDRR)对胶质母细胞瘤细胞增殖、迁移和侵袭的影响,分析其机制是否与调控miR-362-5p表达有关。方法:qRT-PCR检测胶质母细胞瘤组织、细胞以及人脑正常胶质细胞HEB中FENDRR和miR-362-5p表达。以LN229细胞为研究对象,分别构建过表达FENDRR或抑制miR-362-5p表达的LN229细胞株,四甲基偶氮唑蓝(MTT)法检测细胞增殖活力,Transwell实验检测细胞的迁移侵袭能力,Western Blot检测细胞增殖相关蛋白[细胞周期蛋白D1(Cyclin D1)]、迁移侵袭相关蛋白[基质金属蛋白酶2(MMP2)、基质金属蛋白酶9(MMP9)]的表达水平。结果:胶质母细胞瘤组织、细胞中FENDRR的表达水平显著降低,miR-362-5p的表达水平显著升高(P<0.05)。过表达FENDRR或抑制miR-362-5p表达均可抑制Cyclin D1、MMP2和MMP9的表达,抑制LN229细胞的增殖、迁移和侵袭。过表达miR-362-5p可逆转FENDRR对LN229细胞增殖、迁移和侵袭的影响。结论:lncRNA FENDRR可抑制胶质母细胞瘤细胞增殖、迁移和侵袭,其机制可能与靶向负调控miR-362-5p表达有关。 Objective: To investigate the effect of FOXF1 adjacent non-coding developmental regulatory RNA(FENDRR) on the proliferation,migration and invasion of glioblastoma cells by targeting miR-362-5p,and to explore whether its mechanism is related to the regulation of miR-362-5p expression. Methods: The expressions of FENDRR and miR-362-5p in glioblastoma tissues,cells and human brain normal glial cells HEB were detected by qRT-PCR. Taking LN229 cells as the research object,LN229 cell lines overexpressing FENDRR or inhibiting the expression of miR-362-5p were constructed,respectively. MTT assay was used to detect the cell proliferation activity,and Transwell assay was used to detect the cell migration and invasion ability. Western blot was used to detect the expression levels of cell proliferation-related proteins(Cyclin D1) and migration-invasive related proteins(MMP2 and MMP9). Results: The expression of FENDRR was significantly decreased,while the expression level of miR-362-5p was significantly increased in glioblastoma tissues and cells(P<0.05). The over-expression of FENDRR and the inhibition of miR-362-5p both inhibited the expressions of Cyclin D1,MMP2 and MMP9,and inhibited the proliferation,migration and invasion of LN229 cells. Over-expression of miR-362-5p reversed the effects of FENDRR on the proliferation,migration and invasion of LN229 cells. Conclusion: lncRNA FENDRR can inhibit the proliferation,migration and invasion of glioblastoma cells,and the mechanism may be related to the negative targeted regulation of miR-362-5p expression.
