摘要
目的探讨早期姐妹染色单体解离蛋白A(the percocious dissociation of sisters 5A,PDS5A)在胰腺癌中的表达及预后意义。方法应用GEPIA在线工具分析来自TCGA数据库和GTEx项目各种恶性肿瘤及正常组织PDS5A表达情况,筛选具有统计学意义差异表达的恶性肿瘤。同时分析了PDS5A与各恶性肿瘤预后的相关性,筛选有统计学意义预后相关性的癌种。分析PDS5A的表达与胰腺癌分期及预后的关系。从TCGA数据库下载胰腺癌(RNA-seq)基因表达数据,进一步分析与PDS5A基因表达相关的差异基因,进行GO富集及KEGG富集分析。采用Western blot法检测PCNA-1(人胰腺癌细胞)、BON-1(人胰腺神经内分泌瘤细胞)、HPDE6-C7(胰腺导管上皮细胞)中PDS5A表达。结果GEPIA分析显示PDS5A基因在胰腺癌中高表达,差异具有统计学意义,P<0.05。并且PDS5A高表达与胰腺癌患者的OS具有负相关性,P<0.05。富集分析表明PDS5A的高表达与AQP5、SPINK1、MUC6、TFF2等基因负表达有显著相关性,P<0.05。Western blot初步证实胰腺癌细胞中PDS5A表达高于正常胰腺癌细胞。结论PDS5A基因在胰腺癌中高表达,且与患者预后显著相关,可能有望成为胰腺癌判断预后的标志物,同时可能成为治疗靶点。
Objective To investigate the expression and prognostic significance of GEPIA.Methods Online tool was used to analyze the expression of PDS5A in various malignant tumors and normal tissues from TCGA database and GTEX project.At the same time,the correlation between PDS5A and the prognosis of various malignant tumors was analyzed,and the cancer species with statistically significant prognostic correlation was screened.Objective to analyze the relationship between the expression of PDS5A a and the stage and prognosis of pancreatic cancer.The gene expression data of pancreatic cancer(RNA-seq)was downloaded from TCGA database,and the differential genes related to PDS5A gene expression were further analyzed for go enrichment and KEGG enrichment analysis.Western blot was used to detect the expression of PDS5A in PCNA-1(human pancreatic cancer cells),BON-1(human pancreatic neuroendocrine tumor cells)and HPDE-C7(pancreatic duct epithelial cells).Results GEPIA analysis showed that PDS5A gene was highly expressed in pancreatic cancer,the difference was statistically significant,P<0.05.And the OS of PDS5A patients with pancreatic cancer was correlated,P<0.05.Enrichment analysis showed that the amplification of PDS5A was significantly correlated with the negative expression of AQP5,SPINK1,MUC6,TFF2,P<0.05.Western blot showed that the expression of PDS5A in pancreatic cancer cells was higher than that in normal pancreatic cancer cells.Conclusion PDS5A gene is highly expressed in pancreatic cancer and is significantly correlated with the prognosis of patients.It may be a prognostic marker and a therapeutic target for pancreatic cancer.
