摘要
目的通过构建淫羊藿有效成分的靶点蛋白相互作用网络和活性成分作用靶点的网络,对靶点相关的通路和功能进行分析,研究淫羊藿抗抑郁的作用机制。方法采用Traditional Chinese Medicine Systems Pharmacology(TCMSP)数据库查询获取淫羊藿主要的活性成分,通过drugbank获取各成分的作用靶点,使用Cytoscape软件构建活性成分-作用靶点的网络,利用GeneCards、OMIM、NCBI等数据库查询抑郁症相关的靶点,然后将淫羊藿的靶点与抑郁症靶点进行映射。使用String数据库和Cytoscape软件绘制靶点蛋白相互作用网络,采用DAVID数据库对靶点进行GO及KEGG通路分析。结果经过筛选得到淫羊藿的活性成分21个,主要涉及的作用靶点有188个。网络分析结果表明,淫羊藿发挥抗抑郁药效过程中主要涉及毒性物质反应、金属离子反应、细胞外刺激反应、氧化应激反应等生物过程,通过调节PI3K-Akt signaling pathway、TNF signaling pathway、IL-17 signaling pathway等信号通路来发挥抗抑郁作用。结论本研究结果表明淫羊藿抗抑郁具有多成分-多靶点-多途径的特点,为淫羊藿抗抑郁作用机制的研究提供了新的方法、思路和科学的理论指导。
Objective To study the antidepressant mechanism of Herba Epimedii by constructing the target protein interaction network of active ingredients and the target network of active ingredients.MethodsThe traditional Chinese medicine systems pharmacy(tcmsp)database was used to search for the main active components of Epimedium,and the target of each component was obtained through drugbank.The network of active component targets was constructed by Cytoscape software.The depression related targets were searched by genecards,OMIM,NCBI and other databases,and then the targets of Epimedium were mapped to depression targets.String database and Cytoscape software were used to draw the target protein interaction network,and David database was used to analyze the go and KEGG pathways.Results 21 active components of Epimedium were screened,and 188 targets were involved.The results of network analysis showed that the antidepressant effect of Epimedium mainly involved in toxic substance reaction,metal ion reaction,extracellular stimulation reaction,oxidative stress reaction and other biological processes.The antidepressant effect was exerted by regulating PI3 K Akt signaling pathway,TNF signaling pathway,IL-17 signaling pathway and other signaling pathways.Conclusion The results of this study show that the antidepressant effect of Epimedium has the characteristics of multi-component,multi-target and multi-channel,which provides a new method,idea and scientific theoretical guidance for the study of antidepressant mechanism of Epimedium.
