摘要
以热研7-33-97橡胶树组织培养材料为试验材料,通过分子鉴定的方法确认橡胶树组织培养中常见污染源是红青霉菌。研究不同浓度植物组织抗菌剂PTC_(3)(0.2、0.4、0.6、0.8、1、1.2、1.4、1.6、1.8、2.0mL/L)和PPM (0.5、0.55、0.6、0.65、0.7、0.75、0.8、0.85、0.9、0.95 mL/L)抑菌剂对红青霉菌的生长抑制率,及其对橡胶树次生体胚发生效率的影响,结果表明,当PTC_(3)抑菌剂浓度为1 mL/L时,对红青霉菌有一定的抑制作用,培养2 d时和培养5 d时,对真菌的生长抑制率最高分别为18.1%、22.3%。当PPM抑菌剂浓度为0.6、0.65、0.7、0.85、0.95 mL/L时,培养2 d,对真菌的生长抑制率分别为18.6%、17.4%、17.2%、19.2%、16.3%。培养5 d时,所有抑菌剂浓度处理对真菌均没有生长抑制作用。同时发现PTC_(3)和PPM对橡胶树次生体胚发生有一定的促进作用,PTC_(3)促进效果稍好一些。
Medium for tissue culture of rubber clone Reyan 7-33-97 was plated for determination of its common contamination source,and the fungus Penicillium rubrum was confirmed to be the common source through molecular identification.The PDAmedium for tissue culture was added with antifungal agent PTC3 at different concentrations(0.2,0.4,0.6,0.8,1,1.2,1.4,1.6,1.8,2.0 mL·L^(-1))and with antifungal agent PPM at different concentrations(0.5,0.55,0.6,0.65,0.7,0.75,0.8,0.85,0.9,0.95 mL·L^(-1))and then inoculated with the strains of P.rubrum isolated for the culture to observe the growth inhibition rate of the antifungal agents against P.rubrum and their effects on the secondary embryogenesis efficiency of rubber tree.The results showed that the PTC3,when added into the medium at the concentration of 1 mL·L^(-1) had certain inhibitory effect on P.erythraea,and that its growth inhibition rates against P.erythraea were 18.1%and 22.3%,respectively after 2 days and 5 days of culture.The antifungal agent PPM,when supplemented at concentrations of 0.6,0.65,0.7,0.85,0.95 mL·L^(-1) and cultured for 2 days,had the growth inhibition rates of 18.6%and 17.4%,17.2%,19.2%,16.3%,respectively.When cultured for 5 days,all treatments with presence of antifungal agents at all concentrations had no growth inhibitory effect on fungi.At the same time,it was found that PTC3 and PPM had a certain promotion effect on secondary somatic embryogenesis of rubber tree,and that the PTC3 had slightly better promotion effect.
