硫炔丙基半胱氨酸对三种牙周致病菌抗菌作用 被引量:2

S-propargyl-cysteine has antibacterial effect on three periodontal pathogens

摘要 目的:研究硫炔丙基半胱氨酸(S-propargyl-cysteine,SPRC)溶液对牙周炎常见致病菌的体外抗菌作用。方法:体外分别将牙龈卟啉单胞菌(P.gingivalis),中间普氏菌(P.intermedia)和粪肠球菌(E.faecalis)与不同浓度SPRC(10,20,50和100 mmol/L)共培养,没有给药处理的作为对照组(0 mmol/L组),给予2%醋酸氯己定(Chlorhexidine acetate,CHA)处理作阳性对照组;每间隔1 h测定1次各浓度组共培养后的吸光度(OD600nm)值,并绘制10 h生长曲线;采用直接接触法(direct contact test,DCT)和CCK-8法测定不同浓度下共培养5 h后各组吸光度值;采用菌落形成实验(colony-forming units,CFU)检测37℃缺氧条件培养24 h后不同浓度组对菌落数形成的差异。结果:SPRC对E.faecalis生长有影响,在5~10 h内不同浓度给药组与对照组(0 mmol/L组)相比细菌数量明显减少(P<0.05);DCT,CCK-8和CFU法结果均显示SPRC对E.faecalis具有抑菌作用,不同浓度给药组与对照组(0 mmol/L组)相比具有明显差异(P<0.05),且高浓度抑制作用更明显。DCT,CCK-8和CFU法结果显示不同浓度SPRC对P.gingivalis、P.intermedia无抗菌性,均无统计学差异(P>0.05)。结论:体外研究发现SPRC对E.faecalis具有抗菌性,且效果随浓度增加而增强,但对P.gingivalis和P.intermedia无抑制作用。 Objective:To investigate the antibacterial effect of S-propargyl-cysteine(SPRC)solution on common pathogenic bacteria of periodontitis in vitro.Methods:Different concentrations of SPRC(10,20,50,100 mmol/L)was co-cultured with Porphyromonas gingivalis(P.gingivalis),Porphyromonas intermedia(P.intermedia)and Enterococcus faecalis(E.faecalis)in vitro.Untreated group is served as control group(0 mmol/L)and 2%CHA treated group is served as the positive control group.The absorbance(OD600 nm)of each co-cultured concentration group was measured per hour and the total growth curve to 10 hours was analyzed.Direct contact method(DCT)and CCK-8 method were used to measure the absorbance of each group after co-cultivation in different concentrations for 5 hours.Colony formation experiment(CFU)was used to detect the difference in the number of formed colonies after 24 hours culture at 37°C under hypoxic conditions in different concentration groups.Results:SPRC significantly affects the growth of E.faecalis and the number of bacteria which was significantly lower in the SPRC groups(10,20,50,100 mmol/L)than the control group(0 mmol/L)from 5 to 10 hours(P<0.05).DCT,CCK-8 and CFU results showed that SPRC groups had bacteria-inhibiting effect to E.faecalis comparied with control group(0 mmol/L)(P<0.05)and the higher concentrations of SPRC groups would have the better antibacterial effect.However it showed no antibacterial effect on P.gingivalis and P.intermedia(P>0.05).Conclusion:SPRC has antibacterial activity against E.faecalis which was positively enhanced with the increased concentration,but has no inhibitory effect on P.gingivalis and P.intermedia in vitro.
