沙地云杉胚性愈伤组织的褐化因素研究 被引量:2

STUDY ON BROWNING FACTORS OF EMBRYOGENIC CALLUS OF Picea Mongolica ON SANDY LAND

摘要 本论文以成熟胚为外植体,对沙地云杉胚性愈伤组织褐化的主要因素进行了研究,研究不同蔗糖浓度、植物凝胶含量、pH值及肌醇含量对沙地云杉胚性愈伤组织诱导的影响。结果表明:5种蔗糖浓度中,20 g/L时胚性愈伤组织的褐化率较小,诱导率最好,并且胚性愈伤组织长的很好;pH值6种梯度中,在5.75(5.8期间胚性愈伤组织褐化率较小,其诱导率最好);沙地云杉胚性愈伤诱导的培养基中添加2.5 g/L植物凝胶和0.2 g/L肌醇时能保证较低的褐化率,最高的诱导率。通过本研究的调整和优化,在2周内,诱导沙地云杉的胚性愈伤组织时平均褐化率降低到5%左右,不但未影响到后续胚性愈伤组织的增殖效率,而且能达到预期目的。并且解决了沙地云杉成熟胚胚性愈伤组织诱导中存在的愈伤组织诱导率低、生长状态差、褐化率较高等问题,为建立沙地云杉体细胞胚胎发生体系奠定了基础。 In this paper,mature embryos were used as explants to study the main factors affecting the browning of the embryogenic callus of Picea mongolica on the sandy land.The effects of sucrose concentration,plant gel content,pH value and inositol content on the P.mongolica embryogenic callus induction were studied.The results showed that,among the five sucrose concentrations,the browning rate of embryogenic callus was lower at 20 g/L,the induction rate was the best,and the embryogenic callus grew well.Among the six pH gradients,the browning rate of embryogenic callus was lower during the period from 5.75 to 5.8,and the induction rate was the best.A lower browning rate and the highest induction rate values were obtained when the medium for embryogenic callus induction of P.mongolica added 2.5 g/L plant gel and 0.2 g/L inositol.Through the adjustment and optimization of this study,within 2 weeks,the average browning rate of the embryogenic callus of P.mongolica decreased to about 5%,which not only did not affect the proliferation efficiency of subsequent embryogenic callus,but also achieved the desired purpose.The problems of low induction rate,poor growth status,and high browning rate in the embryogenic callus induction of mature embryos of P.mongolica were solved,laying the foundation for establishing the somatic embryogenesis system of P.mongolica.
