miR-106a-5p靶向OSTM1对阿尔茨海默病细胞模型的保护作用

摘要 目的研究miR-106a-5p对阿尔茨海默病(AD)细胞模型存活率和凋亡的影响及潜在分子机制。方法实时荧光定量-聚合酶链式反应(qRT-PCR)检测OSTM1和miR-106a-5p mRNA的表达,噻唑蓝(MTT)法测定AD细胞的存活率,流式细胞术测定AD细胞凋亡率,Western印迹检测OSTM1蛋白表达,双荧光素酶报告系统验证miR-106a-5p与OSTM1的调控关系。结果与对照组相比,模型组miR-106a-5p表达量显著下降(P<0.05),OSTM1 mRNA和蛋白表达量均显著升高(P<0.05);过表达miR-106a-5p和抑制OSTM1表达均可提高AD细胞存活率,抑制AD细胞凋亡;双荧光素酶报告系统结果表明,miR-106a-5p靶向负调控OSTM1的表达;过表达miR-106a-5p会降低OSTM1的表达,抑制miR-106a-5p表达则会提高OSTM1的表达;过表达OSTM1逆转了上调miR-106a-5p对AD细胞存活和凋亡的影响作用。结论miR-106a-5p通过抑制OSTM1表达提高AD细胞存活率,抑制细胞凋亡。miR-106a-5p有望成为AD临床治疗的分子靶点。
