摘要
目的观察电针对慢性限制性损伤(CCI)大鼠海马^(18)F-氟代脱氧葡萄糖(^(18)F-FDG)摄入率及葡萄糖转运体-3(GLUT-3)表达的影响。方法采用随机数字表法将36只雄性SD大鼠分为假手术组、模型组和电针组,每组各12只。模型组和电针组采用4-0铬制羊肠线结扎右侧坐骨神经的方法制备CCI大鼠模型,假手术组只暴露右侧坐骨神经,但不结扎。电针组电针右侧“环跳”“阳陵泉”穴,每次干预30 min。模型组和假手术组只抓取、固定大鼠,不予电针。术后7 d开始,每天1次,连续7 d。术前、干预前后采用Von-Frey针刺痛觉测试套件检测3组大鼠右侧机械痛阈值,干预前后采用Micro PET/CT检测3组大鼠左侧海马^(18)F-FDG摄入率,干预后采用免疫组化检测左侧海马GLUT-3平均光密度。结果干预前与假手术组比较,模型组和电针组机械痛阈值和^(18)F-FDG摄入率均显著下降(P<0.01或0.05);干预后与模型组比较,电针组机械痛阈值和^(18)F-FDG摄入率均显著提高(P<0.01或0.05);与干预前比较,干预后电针组机械痛阈值和^(18)F-FDG摄入率均显著提高(P<0.01或0.05)。干预后与假手术组比较,模型组GLUT-3平均光密度显著下降(P<0.01);干预后与模型组比较,电针组GLUT-3平均光密度显著提高(P<0.01)。结论电针可缓解神经痛大鼠痛敏反应,其机制可能与上调海马GLUT-3表达,提高海马葡萄糖代谢有关。
Objective:To observe the effect of electroacupuncture(EA)on ^(18)F-FDG uptake and GLUT-3 expression in the hippocampus of chronic constriction injury(CCI)rats.Methods:Thirty-six male SD rats were randomly divided into sham group(n=12),model group(n=12)and EA group(n=12).The CCI rat model was established by ligating the right sciatic nerve with 4-0 chromic catgut in the model group and EA group.Rats in the sham group were exposed to the right sciatic nerve without ligation.Rats in the EA group were given EA stimulation on the right side of"Huantiao(GB30)"and"Yanglingquan(GB34)",and each intervention lasted 30 min.Rats in the model group and sham group were only grabbed and fixed,without EA stimulation.All groups were given once a day for 7 consecutive days.Von-Frey test was used to assess the right side of mechanical pain threshold.Micro PET/CT was performed to evaluate the ^(18)F-fluorodeoxyglucose(^(18)F-FDG)uptake in left hippocampus.Immunohistochemistry was used to measure the average optical density of glucose transporter 3(GLUT-3).Results:Before the intervention,the mechanical pain threshold and ^(18)F-FDG uptake of the model group and EA group were obviously reduced than those of the sham group(P<0.01 or P<0.05).After the intervention,the mechanical pain thresholds and ^(18)F-FDG uptake of the EA group were obviously increased than those of the model group(P<0.01 or P<0.05).Compared with before intervention,the mechanical pain thresholds and ^(18)F-FDG uptake of the EA group were obviously increased after intervention(P<0.01 or P<0.05).Compared with the model group,the average optical density of GLUT-3 significantly decreased in sham group after the intervention(P<0.01),and that in the EA group was significantly increased(P<0.01).Conclusions:EA can alleviate hyperalgesia in rats with neuralgia,and its mechanism may be related to up regulating the expression of GLUT-3 in hippocampus and improving glucose metabolism in hippocampus.
