摘要
为了将中药双黄连所含化学成分和其药效相关联,建立能够通过色谱指纹图谱实现准确预测药效的质量控制和评价新模式,实现双黄连的全面、系统的质量控制.本文通过对双黄连样品的前处理方法、色谱指纹图谱和谱效关系进行归纳总结,并对其发展前景进行展望.结果表明:不同剂型双黄连前处理通常用甲醇/水溶剂溶解,其中粒径大的片剂和颗粒剂需经研细后再溶解,经微孔滤膜过滤后待测.色谱指纹图谱技术可以快速、准确地测定双黄连中绿原酸、黄芩苷和连翘苷等有效成分的含量以及重金属、有机氯农药等有害物质的含量.双黄连的谱效关系已证实了双黄连与H1N1流感病毒致犬肾(MDCK)细胞损伤的保护作用和退热作用有相互联系.本文可对后续建立双黄连质量控制和评价新模式提供参考方法和依据.
In order to correlate the chemical composition of Shuanghuanglian with its efficacy,a new mode of quality control and evaluation which can accurately predict the efficacy of Shuanghuanglian through chromatographic fingerprint is established to realize comprehensive and systematic quality control of Shuanghuanglian.In this paper,the sample pretreatment methods,the study of chromatographic fingerprint and spectral-effect relationships were summarized,and their development prospects were predicted.The results showed that the pretreatment of Shuanghuanglian in different dosage forms were usually dissolved by methanol/water,in which tablets and granules with large particle size should be ground and then dissolved,and then filtered by microporous membrane before chromatographic analysis.Chromatographic fingerprint technology could quickly and accurately determine the contents of chlorogenic acid,baicalin,forsythin and other effective components in Shuanghuanglian,as well as the contents of harmful substances such as heavy metals and organochlorine pesticides.The relationship between the spectrum-effect of Shuanghuanglian and the protective effect of Madin-Darby canine kidney(MDCK)cells caused by H1 N1 influenza virus and antipyretic effect has been confirmed.This paper is hoped to provide the method and basis for establishing the new mode of quality control and evaluation of Shuanghuanglian.
