Aha1基因过表达对黑色素瘤细胞凋亡的影响

摘要 目的研究Aha1基因在恶性黑色素瘤形成过程中的作用。方法采用定量PCR和免疫印迹的方法检测并比较小鼠B16F10移植恶性黑色素瘤及瘤旁的组织中Aha1基因的表达。在恶性黑色素瘤B16F10细胞中转染转染真核表达载体过表达Aha1基因后,使用CCK-8法检测细胞的增殖,流式细胞术检测细胞的凋亡。结果Aha1在恶性黑色素瘤中的表达显著低于相应的瘤旁组织,过表达的Aha1基因导致黑色素瘤细胞的生长停滞,并促进黑色素瘤细胞B16F10的凋亡。结论低表达的Aha1基因有利于恶性黑色素瘤的形成。 Objective To study the role of Aha1 gene in the formation of malignant melanoma.Methods Aha1 expression in B16F10 tumor xenograft and peritumoral tissues was determined by quantitative PCR and western blot assays.After Aha1 was over-expressed in B16F10 cells by transfecting with eukaryotic expression vector,cell proliferation was detected by CCK-8 assay,and apoptosis was detected by flow cytometry.Results Aha1 expression in malignant melanoma was significantly lower than that in peritumoral tissues.Aha1 overexpression led to the stagnation of melanoma cell growth and promoted melanoma B16F10 cells apoptosis.Conclusion Low expression of Aha1 is beneficial to the formation of malignant melanoma.
