摘要
利用微波辅助提取,结合苯酚-硫酸法测定魔芋总多糖含量,以总多糖提取率为考察指标,选取微波功率、提取时间、料液比、提取次数为考察因素,在单因素实验的基础上,采用L_(9)(3^(4))正交实验优选最佳提取工艺。综合考虑最佳提取工艺微波功率为560 W、提取时间为30 min、提取料液比1:250、提取次数为2次。验证性实验总多糖平均提取率16.07%,优于传统的水浴提取法,为魔芋药材中有效成分的提取提供了新的方法。
Microwave-assisted extraction and phenol-sulfuric acid method were used to determine the total polysaccharide content of konjac.The total polysaccharide extraction rate was used as the inspection index.Microwave power,extraction time,material-to-liquid ratio,and extraction times were selected as the inspection factors.On the basis of single factor inspection,L_(9)(3^(4))orthogonal test was used to optimize the best extraction process.Considering the optimal extraction process,the microwave power was 560 W,the extraction time was 30 min,the extraction material-liquid ratio was 1:250,and the extraction times were 2 times.The average extraction rate of total polysaccharides in the confirmatory experiment was 16.07%,which was better than the traditional water bath method,and it provided a new method for the extraction of effective ingredients in konjac medicinal materials.
