摘要
以乙酰半胱氨酸为研究对象,采用紫外分光光度法,选用Ellman试剂来检测乙酰半胱氨酸含量,研究发现淡黄色的Ellman试剂(DTNB)将转变成黄色的5-巯基-2-硝基苯甲酸,5-巯基-2-硝基苯甲酸在412 nm处具有最大吸收,且DTNB的吸收光谱并不干扰巯基的测定,具有操作简单,检测费用少,分析速度快,稳定性良好,灵敏度高的特点。为乙酰半胱氨酸工业化测定提供了可行性方案。
Acetylcysteine was used as the research object,and ultraviolet spectrophotometry was used to detect the content of acetylcysteine using Ellman reagent.It was found that the light yellow Ellman reagent(DTNB)would be converted into yellow 5-mercapto group.5-mercapto-2-nitrobenzoic acid had the maximum absorption at 412nm,and the absorption spectrum of DTNB does not interfere with the determination of sulfhydryl groups.It had simple operation,low detection cost,fast analysis speedand stability,featured good performance and high sensitivity.It provided a feasible scheme for the industrial determination of acetylcysteine.
