超声波辅助纤维素酶提取金荞麦总黄酮工艺优化及其抗氧化活性研究 被引量:11

Ultrasound-assisted cellulase optimization of extraction process of total flavonoids from Fagopyrum dibotrys(D.Don)Hara and its antioxidant activity research

摘要 以金荞麦为原料,采用单因素试验和响应面法优化超声波辅助纤维素酶提取金荞麦总黄酮的工艺。探讨了乙醇体积分数、加酶量、酶解时间、酶解温度对金荞麦中总黄酮提取率的影响,并对金荞麦总黄酮的体外抗氧化活性进行评价。结果表明:最佳提取工艺参数为乙醇体积分数60%、加酶量11 mg、酶解时间105 min、酶解温度64℃,在此条件下,金荞麦总黄酮的提取率为14.25%;扫描电镜观察发现,超声辅助酶法处理对金荞麦微观结构破坏程度较大;金荞麦总黄酮DPPH自由基清除率为86.09%,具有较好的抗氧化活性。 The technology of ultrasonic-assisted cellulase extraction of total flavonoids from Fagopyrum dibotrys(D.Don)Hara was optimized by single factor experiment and response surface method with Fagopyrum dibotrys(D.Don)Hara as the raw material.The effects of ethanol volume fraction,amount of enzyme added,enzymolysis time and temperature on the extraction rate of total flavonoids in Fagopyrum dibotrys(D.Don)Hara were discussed,and the in vitro antioxidant activity of total flavonoids in Fagopyrum dibotrys(D.Don)Hara was evaluated.The results showed that the optimal extraction process parameters were as follows:ethanol volume fraction 60%,enzyme added amount 11 mg,enzymolysis time 105 min,enzymolysis temperature 64℃.Under these conditions,the extraction rate of total flavonoids from Fagopyrum dibotrys(D.Don)Hara was 14.25%;Scanning electron microscope observation revealed that ultrasonic-assisted enzymatic treatment had a greater degree of damage to the microstructure of Fagopyrum dibotrys(D.Don)Hara;The scavenging rate of DPPH free radical of total flavonoids from Fagopyrum dibotrys(D.Don)Hara was 86.09%,which had good antioxidant activity.
