细粒棘球蚴脂肪酸转运蛋白生物学特性分析 被引量:1

摘要 本研究根据NCBI GenBank数据库中的Eg-FATP氨基酸序列,采用生物信息学方法分析细粒棘球蚴脂肪酸转运蛋白(Eg-FATP)理化性质,亚细胞定位、抗原表位、信号肽、跨膜区二级和三级结构、系统进化树、蛋白互作网络等,并通过qPCR方法探究其在原头蚴和包囊壁的差异表达,旨在为后期相关的研究提供理论支持和数据参考。结果显示,Eg-FATP蛋白无信号肽,无跨膜区,亚细胞定位于细胞质;该蛋白有10个B细胞线性表位和14个CTL细胞表位;系统发育分析显示:细粒棘球蚴的FATP蛋白与多房棘球蚴亲缘关系较近,细粒棘球蚴中国株和英国株之间的氨基酸序列同一性较高,为97%左右。预测的互作功能蛋白可能参与胆固醇代谢的调节、脂肪酸跨血屏障的转运等过程;GO和KEGG富集分析结果显示,Eg-FATP对长链和超长链脂肪酸具有酰基辅酶A连接酶活性;qPCR结果显示,FATP基因在原头蚴阶段转录水平较高于包囊壁,具有统计学意义(P<0.05)。本研究对细粒棘球蚴FATP基因的生物学特性进行了初步分析,为进一步揭示其生物学功能奠定了基础。 Based on the amino acid sequence of fatty acid transport protein of Echinococcus granulosus (Eg-FATP) in NCBI GenBank database,the physicochemical properties,subcellular localization,antigenic epitope,signal peptide,transmembrane region,secondary and tertiary structure,phylogenetic tree and protein interaction network of Eg-FATP were analyzed by bioinformatics method along with the differential expression of Eg-FATP in the protocercariae and cyst walls in order to provide theoretical support and data reference for later related research.The results showed that there was no signal peptide and transmembrane region in Eg-FATP protein.The protein had 10 B-cell linear epitopes and 14 CTL cell epitopes.Phylogenetic analysis showed that the Eg-FATP protein was closely related to E.multilocularis.The amino acid sequence identity of E.granulosus between Chinese strain and British strain was about 97%.The predicted interaction functional proteins might be involved in the regulation of cholesterol metabolism and the transport of fatty acids across the blood barriers.Go and KEGG enrichment analysis showed that Eg-FATP had acyl-coa ligase activity for long-chain and ultra-long-chain fatty acids.The results of qPCR revealed that the transcription level of Eg-FATP gene in protoscolices was statistically higher (P< 0.05) than that in cyst walls.The findings of the biological characteristics of the E.granulosus FATP laid the foundation for further research of its biological function.
