摘要
针对荆条转录间隔区设计一套引物,通过温度优化、特异性和灵敏度试验,建立一种荆条蜜源DNA的实时环介导等温扩增(LAMP)检测方法.结果显示,LAMP检测方法在温度为59℃时,引物能够将荆条、椴树、大枣和洋槐DNA区分开,灵敏度可达100 pg·μL^(-1).LAMP方法能对荆条蜜源成分进行快速有效检测,为荆条蜜真实性检测提供一种快速准确的检测方法.
A real-time loop-mediated isothermal amplification(LAMP)method for the detection of DNA from Vitex negundo honey source was established by designing a set of primers for transcription interval.The results showed that the LAMP method could distinguish DNA from brass,linden,jujube and locust at 59℃with a sensitivity of 100 pg·μL^(-1).LAMP method can quickly and effectively detect the source components of Vitex negundo,providing a fast and accurate detection method for the authenticity detection of Vitex negundo.
