摘要
目的探讨PCPA致失眠大鼠的时钟基因表达变化。方法雄性SD大鼠一次性腹腔注射PCPA复制失眠动物模型,采用旷场实验评价失眠大鼠的行为学变化,采用ELISA法检测外周血清中HPA轴激素水平,采用RT-qPCR技术检测大鼠脑部、肝脏核心时钟基因(Clock、Baml1、Per1、Per2、Cry1和Cry2)的转录水平。结果与对照组比较,模型组大鼠行为学得分显著升高(P<0.01);脑部Clock、Bmal1、Cry1表达下调(P<0.05),Cry2的表达显著上升(P<0.05);肝脏Per1表达上升(P<0.01)。结论PCPA诱导的大鼠失眠可能与脑部Clock、Bmal1、Cry1和Cry2基因和肝脏Per1基因的表达改变有关。
Objective To investigate the changes of clock gene expressions of brain and liver in the insomnia rats induced by PCPA.Methods Male SD rats were intraperitoneally injected with PCPA to establish the insomnia animal model.The behavioral changes of insomnia rats were evaluated by open field experiment.The levels of HPA axis hormones in peripheral serum were detected by ELISA.The transcription levels of core clock genes(Clock,Baml1,Per1,Per2,Cry1 and Cry2)in brain and liver of rats were detected by RT-qPCR.Results Compared with the control group,the behavioral score of rats in the model group was significantly higher(P<0.01).The expressions of Clock,Bmal1 and Cry1 in brain were down-regulated(P<0.05).Moreover,the expression of Per1 in the liver was up-regulated(P<0.01).Conclusion PCPA-induced insomnia in rats may be associated with altered expressions of Clock,Bmal1,Cry1 and Cry2 genes in the brain and Per1 genes in the liver.
