摘要
目的基于网络药理学方法探讨柴胡治疗动脉粥样硬化的有效活性成分及靶点的作用机制。方法该研究运用TCMSP数据库结合筛选条件确定柴胡的主要有效活性成分及其作用靶点,构建主要有效活性成分-靶点网络图。利用GeneCards数据库获取动脉粥样硬化疾病靶点,取柴胡和动脉粥样硬化的交集靶点,对交集靶点进行蛋白质互相作用网络分析、基因功能富集分析和信号通路富集分析,并对药物-主要有效活性成分-疾病-靶点进行网络分析。结果筛选主要有效活性成分17种,其中6个有效活性成分没有相对应靶点,并获得药物靶点93个。通过GeneCards数据库,获得1217个疾病靶点,药物与疾病交集基因共57个,PPI蛋白互作网络分析得出MAPK8、IL6、VEGFA、EGFR、ESR1、CASP3、MYC、CCND1等可能是柴胡治疗动脉粥样硬化的核心靶点。GO功能富集分析得到219个生物过程条目,50个细胞组成条目,23个分子功能条目。KEGG通路富集分析排名靠前的通路主要有乙型肝炎、癌症相关的通路、癌症中的蛋白多糖、结直肠癌、胰腺癌、TNF信号通路等。结论柴胡治疗动脉粥样硬化具有多途径、多靶点的作用特点,为探析中医治疗动脉粥样硬化提供参考。
Objective To explore the effective active components and target mechanism of Chaihu(Bupleuri Radix)in the treatment of atherosclerosis based on network pharmacology. Methods This study used TCMSP database combined with screening conditions to determine the main effective active ingredients and their targets,and constructed the main effective active ingredient-target network graph. The target of atherosclerosis disease was obtained by using GeneCards database,the intersection target of Chaihu(Bupleuri Radix)and atherosclerosis was taken,protein interaction network analysis,gene function enrichment analysis and signal pathway enrichment analysis were performed on the intersection target,and the drug-main active ingredient-disease-target network analysis was conducted. Results 17 main active ingredients were screened,among which 6 active ingredients had no corresponding target,and 93 drug targets were obtained. Through GeneCards database,1217 disease targets were obtained,57 genes of drug and disease intersection were obtained,PPI protein interaction network analysis showed that MAPK8,IL6,VEGFA,EGFR,ESR1,CASP3,MYC,CCND1,etc. May be the core targets of Chaihu(Bupleuri Radix)for the treatment of atherosclerosis. GO functional enrichment analysis yielded 219 biological processes,50 cell components and 23 molecular functions. KEGG pathway enrichment analysis ranked the highest in hepatitis B,cancer-related pathways,proteoglycan in cancer,colorectal cancer,pancreatic cancer,TNF signaling pathway,etc. Conclusion Chaihu(Bupleuri Radix)has the characteristics of multi-pathway and multi-target in the treatment of atherosclerosis.
