摘要 目的:观察大黄对大鼠肠道菌群的影响,以积累大黄在临床实践中合理使用的数据。方法:对普通大鼠分别灌予低、中、高剂量的大黄,分别在连续给药时间7、14和21天等时间点收集大鼠十二指肠、空肠、回肠和结肠等肠段(含肠内容物)样本,然后通过实时荧光定量PCR(qRT-PCR)技术检测各肠段大肠杆菌、乳酸菌、拟杆菌和梭菌等4种肠道主要优势菌的数量。结果:(1)大黄对十二指肠大肠杆菌的生长具有抑制效应,对回肠大肠杆菌的生长具有促进效应,对空肠大肠杆菌生长的影响是先抑制后促进,而对结肠大肠杆菌生长的影响是先促进后抑制;(2)大黄对空肠、回肠和结肠拟杆菌的生长具有促进效应,而对十二指肠拟杆菌生长的影响是先促进后抑制;(3)大黄对十二指肠乳酸菌的生长具有促进效应,对空肠乳酸菌生长的影响是先抑制后促进,对回肠和结肠乳酸菌生长的影响是先促进后抑制;(4)大黄对十二指肠、空肠、回肠和结肠梭菌的生长具有抑制效应。以上促进或抑制效应在各肠段有一定的差异,且存在量-时规律。结论:大黄对肠道菌群的影响在各肠段存在一定的差异,且具有明显的量-时规律。 Objective:To observe the effect of rhubarb on the intestinal flora of rats in order to accumulate data on the rational use of rhubarb in clinical practice.Methods:Ordinary rats were perfused with low,medium and high doses of rhubarb,and the duodenum,jejunum,ileum,colon and other intestinal segments of the rats were collected at the time points of the 7 th,the 14 th and the 21 th days of continuous administration(samples containing intestinal contents).Then we detected the number of four major intestinal bacteria,including Escherichia coli,Lactobacillus,Bacteroides and Clostridium,in each segment of the intestine by real-time fluorescent quantitative PCR(qRT-PCR)technology.Results:(1)Rhubarb had an inhibitory effect on the growth of duodenal E.coli and a promotion effect on the growth of ileal E.coli.The effect on the growth of jejunal E.coli was first inhibition and then promotion,while the effect on the growth of colonic E.coli was first promotion and then inhibition;(2)Rhubarb had a promoting effect on the growth of Bacteroides in the jejunum,ileum and colon,while the effect on the growth of Bacteroides duodenum was first promotion and then inhibition;(3)Rhubarb had a promoting effect on the growth of duodenal lactic acid bacteria,while the effect on the growth of jejunum lactic acid bacteria was first inhibition and then promotion,and the effect on the growth of ileum and colon lactic acid bacteria was first promotion and then inhibition;(4)Rhubarb had an inhibitory effect on the growth of duodenum,jejunum,ileum and clostridium colon.The above promoting or inhibiting effects had certain differences in each intestinal segment,and there was a quantity-time rule.Conclusion:The effect of rhubarb on the intestinal flora is different in each intestinal segment,and it has obvious quantity-time rule.
