慢性不可预见应激刺激对小鼠体重的动态变化和炎症因子水平的影响 被引量:2

摘要 目的观察慢性不可预见应激刺激(CUMS)对小鼠的体重动态变化和炎症因子水平的影响。方法取42只雄性BALB/c小鼠随机分为对照组(7只)和模型组(35只),模型组小鼠给予CUMS 4周建立抑郁症小鼠模型。每隔两天监测一次小鼠的体重变化,4周后进行强迫游泳实验、蔗糖偏好实验、旷场实验等行为学实验来确定抑郁症模型是否成功,最后采用酶联免疫吸附实验测定血清前炎症细胞因子(TNF-α、IL-6)和抗炎症细胞因子(IL-10)水平。结果造模第24天后,模型组和对照组小鼠体重开始出现差异(P<0.05);造模第28天后,行为学检测小鼠强迫游泳实验中不动时间、糖水偏好程度、旷场实验中穿越中心次数显示两组小鼠比较差异有统计学意义(P<0.05);同时,前炎症细胞因子TNF-α和IL-6水平较对照组升高,抗炎症细胞因子IL-10较对照组显著降低(P<0.05)。结论CUMS所致BALB/c小鼠抑郁行为可能与炎症因子的异常表达有关,同时抑郁模型小鼠体重差异最早可在造模24 d后开始出现,且此差异随时间延长差异越显著,这一发现为抑郁症的早期发现、治疗和干预提供实验基础研究。 Objective To observe the impacts of chronic unpredictable stress(CUMS)on dynamic changes of body weight and inflammatory factors in mice.Methods A total of 42 male BALB/c mice were selected and randomly divided into the control group(n=7)and the model group(n=35).Mice in the model group were treated withCUMS for 4 weeks to establish a depression mice model.The weight changes of mice were monitored every 2 days.After 4 weeks,behavioral experiments such as forced swimming test,sucrose preference test and open field test were conducted to determine whether the depression model was successful.Finally,the levels of serum pro-inflammatory cytokines(TNF-α,IL-6)and anti-inflammatory cytokines(IL-10)were measured by enzyme-linked immunosorbent assay.Results On the 24th day after modeling,the weight of mice in the model group and the control group began to show difference(P<0.05).On the 28th day after modeling,behavioral test showed that there were significant differences between the two groups in immobility time in forced swimming test,sucrose preference test and times of crossing center in open field test(P<0.05).Meanwhile,the levels of pro-inflammatory cytokines TNF-αand IL-6 were higher than those of the control group,and anti-inflammatory cytokine IL-10 was significantly lower than that of the control group(P<0.05).Conclusion CUMS-induced depression in BALB/c mice may be related to the abnormal expression of inflammatory factors,and the weight difference of depression model mice can begin to occur as early as 24 days after modeling,and the difference is more significant with time.This discovery provides experimental basis for early detection,treatment and intervention of depression.
