摘要
目的通过检测不同浓度IL-17处理后宫颈癌Caski细胞与人外周血单核细胞(PBMC)共培养系统下癌细胞生长率及凋亡率情况,探讨IL-17所介导的免疫反应在高危型HPV相关宫颈癌发展中的作用。方法选取2017年9月至2019年6月期间我院就诊的健康女性及部分本院女性志愿者,抽取外周抗凝血10 mL用于提取人外周血单核细胞(PBMC),与包含高危型HPV序列的宫颈癌Caski细胞株进行共培养,经过不同浓度的rhIL-17处理,采用CCK-8法检测各组癌细胞的生长率,采用流式细胞术检测各组癌细胞的凋亡率。结果①CCK-8法结果显示,低浓度组癌细胞生长率最高,为(125.65±7.28)%,高于对照组,差异有统计学意义(P<0.05);高浓度组癌细胞生长率为(110.56±9.39)%,同样高于对照组,低于低浓度组,但差异无统计学意义(P>0.05);②流式细胞术结果显示,低浓度组宫颈癌细胞的凋亡率最低,为(10.67±0.20)%,低于对照组的(12.43±0.81)%,差异有统计学意义(P<0.05);高浓度组癌细胞的凋亡率为(11.99±0.11)%,同样低于对照组,高于低浓度组,但差异无统计学意义(P>0.05)。结论IL-17所介导的免疫反应与宫颈癌的发展密切相关,其在肿瘤发展的不同阶段表现出双重效应,为进一步研究高危型HPV相关宫颈癌的免疫治疗措施提供了基本理论依据。
Objective To detect the growth rate and apoptosis rate of cancer cells in the co-culture system of Caski cells in cervical cancer and human peripheral blood mononuclear cells(PBMC)treated with different concentrations of IL-17,so as to investigate the function of IL-17 mediated immune reaction in the development of high-risk HPV-related cervical cancer.Methods Healthy women and some female volunteers admitted to our hospital from September 2017 to June 2019 were selected,and 10 mL of peripheral anticoagulant was used to extract human PBMC,which were co-cultured with Caski cell in cervical cancer containing high-risk HPV sequence.After treatment with different concentrations of rhIL-17,the growth rate of each group of cancer cells was detected by CCK-8 method,and the apoptosis rate of each group of cancer cells was detected by flow cytometry.Results(1)CCK-8 method showed that the growth rate of cancer cells in the low concentration group was the highest(125.65±7.28)%,which was higher than that in the control group,with statistically significant difference(P<0.05);The growth rate of cancer cells in the high concentration group was(110.56±9.39)%,which was also higher than that in the control group,the growth rate of cancer cells in the high concentration group was lower than that in the low concentration group,but without statistically significant difference(P>0.05).(2)The results of flow cytometry showed that the apoptosis rate of cervical cancer cells in the low concentration group was the lowest(10.67±0.20)%,which was lower than that of(12.43±0.81)%in the control group,with statistically significant difference(P<0.05);The apoptosis rate of cancer cells in the high concentration group was(11.99±0.11)%,also lower than that in the control group,the apoptosis rate of cervical cancer cells in the high concentration group was higher than that in the low concentration group,but without statistically significant difference(P>0.05).Conclusion The immune reaction mediated by IL-17 is closely related to the development of cervical cancer,and it shows double effects in different stages of tumor development,which provides a basic theoretical basis for further research of immunotherapy measures for high-risk HPV-related cervical cancer.
