聚乙烯醇联合藻酸钙与兔软骨细胞共培养对兔膝关节软骨缺损修复的影响 被引量:1

摘要 目的探讨聚乙烯醇联合藻酸钙(PVA/CaAlg)与兔软骨细胞(ATDC-5)共培养对兔膝关节软骨缺损修复的影响,以及对胰岛素样生长因子-1(IGF-1)的mRNA表达及Ⅱ型胶原分泌的影响。方法将20只成年新西兰兔随机分为实验组与对照组,每组10例,在右膝关节软骨造缺损模型。将从4周龄新西兰兔膝关节组织中收集的ATDC-5细胞制成细胞悬液,并经培养传代后接种于PVA/CaAlg复合物上。混合培养2周后,将PVA/CaAlg复合物植入实验组的兔造膝关节缺损模型中;对照组缺损区不做处理。结果PVA/CaAlg复合物内ATDC-5细胞活性良好;植入缺损关节12周后,实验组缺损区ATDC-5细胞数量、Ⅱ型胶原的分泌以及IGF-1的mRNA表达量均高于对照组(P<0.05)。结论将PVA/CaAlg复合物与ATDC-5细胞共培养物植入兔膝关节软骨缺损区,能促进Ⅱ型胶原的分泌和IGF-1的mRNA表达,改善缺损区的修复状况。 Objective To investigate the effect of co-culture of polyvinyl alcohol combined with calcium alginate(PVA/CaAlg)and ATDC-5 cells on repairing the articular cartilage defects in rabbits.In additon,the effect of the mRNA expression of insulin-like growth factor-1(IGF-1)and the secretion of typeⅡcollagen.Methods Twenty adult New Zealand rabbits were randomly divided into experimental group and control group,10 cases in each group,and defect model was established in the right knee cartilage.The ATDC-5 cells collected from the knee tissue of 4-week-old New Zealand rabbits were made into cell suspensions,and cultured and subcultured on the PVA/CaAlg complex.After 2 weeks of mixed culture,the PVA/CaAlg complex was implanted into the rabbit knee joint defect model of the experimental group,and the defect area of the control group was not treated.Results The activity of ATDC-5 cells in PVA/CaAlg comlex was favorable.After 12 weeks of implantation,the number of ATDC-5 cells,the secretion of collagenⅡand the mRNA expression of IGF-1 in the experimental group were higher than those in the control group(P<0.05).Conclusions Implanting co-cultured PVA/CaAlg with ATDC-5 cells into the cartilage defects of rabbit knee joint can promote the secretion of typeⅡcollagen and the mRNA expression of IGF-1,and improve the repair condition of defect area.
