胰岛素联合亚硒酸钠对高糖环境下肾小球系膜细胞增殖的影响

摘要 目的:探讨胰岛素联合亚硒酸钠在高糖环境下对人肾小球系膜细胞(Human Mesangial Cell,HMC)增殖的影响。方法:利用CCK8法检测高糖培养环境下不同剂量的胰岛素或胰岛素+亚硒酸钠处理后HMC的增殖情况。结果:不同浓度(30 mU/mL、100 mU/mL、300 mU/mL)的胰岛素均能显著抑制高糖诱导的HMC异常增殖(P<0.05),其中以100 mU/mL胰岛素的抑制效果最佳;100 mU/mL胰岛素联合500 ng/mL亚硒酸钠后,对高糖诱导HMC增殖的抑制效果更好。结论:亚硒酸钠能增强胰岛素对高糖诱导HMC增殖的抑制效果,提示胰岛素联合亚硒酸钠可能会对糖尿病肾病引起的肾小球硬化具有重要的防治作用。 Objective:To explore the effect of insulin combined with sodium selenite on the proliferation of human mesangial cell(HMC)under high glucose environment.Methods:The CCK8 method was used to detect the proliferation of HMCs induced by high glucose after treatment with different doses of insulin or insulin and sodium selenite combination.Results:Different concentration of insulin(30 mU/mL,100 mU/mL,300 mU/mL)could significantly inhibit the abnormal proliferation of HMC induced by high glucose(P<0.05).Insulin(100 mU/mL)combined with sodium selenite(500 ng/mL)could also significantly inhibit the abnormal proliferation of HMC induced by high glucose(P<0.05),but its effect was better than 100 mU/mL insulin alone(P<0.05).Conclusion:Sodium selenite could enhance the inhibitory effect of insulin on high glucose induced HMC proliferation,suggesting that insulin combined with sodium selenite may has important protect role in attenuating glomerulosclerosis in diabetic nephropathy.
