摘要
目的以藤黄为碳源制备出新型中药衍生碳量子点,探究其抗肿瘤活性。方法通过碱掺杂一步水热法制备藤黄碳量子点,并通过透射电镜、紫外-可见光谱、荧光光谱、X射线光电子能谱、傅里叶红外光谱进行表征,MTT法进行细胞毒性评价。结果碳量子点平均粒径为3.33 nm,分散均匀,量子产率为0.4%,在365 nm紫外灯照射下发出蓝色荧光,紫外-可见光谱和荧光光谱表明藤黄碳量子点具有优异的荧光特性,最大激发波长在367 nm,发射波长在455 nm,细胞毒性实验发现藤黄碳量子点对免疫细胞RAW264.7细胞毒性低,而对Bel 7402肝癌细胞呈现出高抗肿瘤活性。结论成功制备了以藤黄为来源的荧光碳量子点,具有极好的水溶性和优异的光致发光性,通过细胞毒性研究发现该碳量子点可改善原材料藤黄的药理活性,对正常细胞呈现低细胞毒性而对肿瘤细胞表现出高抑制活性,具有一定的抗肿瘤效果,也为后续进一步研究抗肿瘤活性机制提供了一定的基础。
Objective To prepare a new type of carbon quantum dots derived from traditional Chinese medicine gamboge,and determine its anticancer activity.Methods One step hydrothermal method with alkali doping was used to prepare gamboge carbon quantum dots(GCDs),and its characterization and cytotoxicity were evaluated by transmission electron microscopy,UV-Vis spectrum,fluorescence spectrum,X-ray photoelectron spectrum,Fourier infrared spectrum and MTT.Results The average particle size of the prepared carbon quantum dots was 3.33 nm,evenly dispersed,with the quantum yield of 0.4%.The blue fluorescence emitted under the ultraviolet irradiation of 365 nm.UV-vis and fluorescence spectra showed that GCDs had excellent fluorescence characteristics,with the maximum excitation wavelength of 367 nm and emission wavelength of 455 nm.Cytotoxicity experiments found that GCDs showed low cytotoxicity to the immune cells RAW264.7,but highly cytotoxicity to Bel 7402 liver cancer cells.Conclusion GCDs are successfully preparated and with excellent water solubility and photoluminescence properties.GCDs can improve the pharmacological activity of raw material gamboge,showing low cytotoxicity to normal cells while high inhibition activity on cancer cells,and providing reference for further research on the anticancer mechanism.
