摘要
大分子药物如多肽、蛋白质、抗体、聚糖与核酸等在肿瘤、心脑血管疾病和自身免疫疾病等重大疾病的治疗中有极其重要的作用。然而这些大分子药物,由于其分子量大、独特的亲水性和带电状态,通常表现出相对较低的通透性,很难通过皮肤的角质层。近年来,随着对大分子药物促渗方法研究的不断深入,基于物理、化学、纳米载体等方法的生物大分子透皮递送研究取得了很大进展,本文对相关研究进行系统评述。
Macromolecular drugs,such as peptides,proteins,antibodies,glycans and nucleic acids,play an important role in the treatment of major diseases such as tumors,cardiovascular and cerebrovascular diseases and autoimmune diseases.Due to the high molecular weight of these macromolecules,unique hydrophilicity and electric charge state,these macromolecules usually show relatively low permeability,and are difficult to pass through the stratum corneum of the skin.In recent years,with the development in macromolecular drug penetration promotion,great progress has been made in biological macromolecular transdermal delivery based on physical,chemical and nanocarrier methods.This paper systematically reviews related research.
