摘要
目的探究miR-145-5p和SERPINE1在胃癌上皮细胞中的潜在调控机制,希望能够为胃癌的精准治疗提供新的思路。方法从TCGA数据库中下载TCGA-STAD的miRNA成熟体、mRNA和临床数据。利用EdgeR包对mRNA进行差异分析获得DEGs,并与预测基因取交集获得靶标基因。通过qRT-PCR检测miR-145-5p和SERPINE1在正常人胃黏膜上皮细胞和胃癌细胞中的表达情况。通过Western blot检测SERPINE1的蛋白表达情况;通过双荧光素酶实验验证miR-145-5p与SERPINE1之间的靶向关系。用CCK8实验、细胞克隆实验、Transwell法和划痕愈合实验检测转染细胞后细胞的增殖、侵袭与迁移能力。结果MiR-145-5p在胃癌细胞中低表达,SERPINE1在胃癌细胞中高表达,miR-145-5p能够通过调控SERPINE1的表达影响胃癌细胞功能。结论MiR-145-5p在胃癌细胞中发挥重要作用,能够通过靶向下调SERPINE1抑制胃癌细胞增殖、侵袭和迁移。
Objective This study aims to explore the potential regulatory mechanism of miR-145-5p and SERPINE1 in gastric cancer epithelial cells,which can provide new insights for the precise treatment of gastric cancer.Methods Expression profiles of mature miRNAs,mRNAs and clinical data of TCGA-STAD were downloaded from TCGA database.EdgeR package was used to perform differential analysis on mRNAs to obtain differentially expressed genes which were then intersected with predicted genes to obtain target gene.The expression of miR-145-5p and SERPINE1 in normal human gastric mucosal epithelial cells and gastric cancer cells was detected by qRT-PCR.The protein expression of SERPINE1 was detected by western blot.The targeted relationship between miR-145-5p and SERPINE1 was verified by dual luciferase experiment.CCK8,colony formation,transwell and scratch healing assays were used to detect cell proliferative,invasive and migratory abilities after cell transfection.Results MiR-145-5p was lowly expressed,while SERPINE1 was highly expressed in gastric cancer cells.MiR-145-5p could affect the function of gastric cancer cells by regulating the expression of SERPINE1.Conclusion MiR-145-5p plays an important role in gastric cancer cells,which can inhibit the proliferation,invasion and migration of gastric cancer cells by targeting and down-regulating SERPINE1.
