摘要
目的:探讨右美托咪定对结直肠癌LOVO细胞侵袭和迁移的影响及其发生机制。方法:以0、50、100和200μg/mL右美托咪定处理48 h后,采用细胞计数(CCK-8)法检测细胞活力,Transwell小室检测细胞侵袭和迁移能力,实时荧光定量聚合酶链反应(PCR)法检测细胞中微小RNA(miR)-196a-5p的表达水平。将miR-196a-5p抑制剂转染至LOVO细胞后,观察下调miR-196a-5p表达对右美托咪定作用下的LOVO细胞活力、侵袭和迁移的影响。结果:与0μg/mL组比较,50、100和200μg/mL组右美托咪定可增强LOVO细胞活力、促进细胞侵袭和迁移并上调细胞中miR-196a-5p表达水平(P<0.05),且呈现一定的浓度依赖性。转染miR-196a-5p抑制剂后,右美托咪定对LOVO细胞活力、细胞侵袭和迁移的促进作用明显减弱(P<0.05)。结论:右美托咪定可通过上调miR-196a-5p表达促进结直肠癌LOVO细胞侵袭和迁移。
Objective:To investigate the effect and mechanism of dexmedetomidine on the invasion and migration of LOVO cells in colorectal cancer.Methods:After treatment with 0,50,100 and 200μg/ml dexmedetomidine for 48 hours,the cell viability was detected by CCK-8 method,the cell invasion and migration ability was detected by Transwell cell chamber,and the expression level of microRNA(miR)-196a-5p was detected by real-time fluorescence quantitative PCR.After miR-196a-5p inhibitor was transfected into LOVO cells,the effects of down-regulation of miR-196a-5p expression under the action of dexmedetomidine on the viability,invasion and migration of LOVO cells were observed.Results:Compared with the 0μg/ml group,50,100 and 200μg/ml dexmedetomidine could enhance the activity of LOVO cells,promote cell invasion and migration,and up regulate the expression of miR-196a-5p in the cells(P<0.05).After transfection with miR-196a-5p inhibitor,the promoting effect of dexmedetomidine on LOVO cell viability,cell invasion and migration decreased significantly(P<0.05).Conclusion:Dexmedetomidine can promote the invasion and migration of LOVO cells in colorectal cancer by up regulating the expression of miR-196a-5p.
