子宫颈癌及病变中信号通路Shh蛋白与p-STAT3的表达及临床意义 被引量:1

摘要 目的探讨音刺猬蛋白(Shh)、磷酸化信号传导与转录活化因子-3(p-STAT3)在宫颈癌发生、发展中的作用。方法应用免疫组织化学检测2015年1月至2018年12月该院收治的30例正常宫颈(NC)患者、35例低级别鳞状上皮内病变(LSIL)患者、35例高级别鳞状上皮内病变(HSIL)患者及45例宫颈鳞状细胞癌(SCC)患者宫颈组织中Shh和p-STAT3的表达,并进行数据分析。结果Shh、p-STAT3的表达随肿瘤进展而增强。Shh、p-STAT3在NC、SIL组及SCC各组间阳性表达率比较,差异均有统计学意义(P<0.05)。LSIL组与SCC组间、HSIL组与SCC组间Shh阳性表达率比较,差异均有统计学意义(P<0.05);LSIL组与SCC组间、HSIL组与LSIL组间p-STAT3阳性表达率比较,差异均有统计学意义(P<0.05),LSIL组与NC组、HSIL组与SCC组p-STAT3阳性表达率比较,差异均无统计学意义(P>0.05)。在宫颈癌组织中,Shh和p-SATA3的表达与肿瘤的临床分期、肿瘤分化程度及淋巴结转移有关(P<0.05),和肿瘤大小及年龄无关(P>0.05)。癌组织中p-STAT3的表达与Shh的表达关系密切(r=0.448,P<0.05)。结论Shh信号途径及STAT3信号途径与宫颈癌的发生及进展有一定的关系,并且2种信号途径的活化有协同作用。 Objective To explore the role of sonic hedgehog(Shh),phosphorylation signal transduction and activator of transcription-3(p-STAT3)in the occurrence and development of cervical cancer.Methods The expression of Shh and p-STAT3 in 30 normal cervical(NC),35 low-grade squamous intraepithelial lesions(LSIL),35 high-grade squamous intraepithelial lesions(HSIL)and 45 cervical squamous cell carcinoma(SCC)tissues of patients who were admitted to the hospital from January 2015 to December 2018 were detected by immunohistochemistry,and the data was analysised.Results The positive expression rate of Shh and p-STAT3 were enhanced with tumor progression.The positive expression rate of Shh、p-STAT3 were statistically significant between NC、SIL and SCC groups(P<0.05).The difference in the positive expression rate of Shh between the LSIL group and the SCC group,and between the HSIL group and the SCC group were statistically significant(P<0.05).The positive expression rate of p-STAT3 were statistically significant in LSIL and SCC groups compared with HSIL and LSIL groups(P<0.05),the positive expression rat of p-STAT3 were no significant differences between LSIL group and NC group,HSIL group and SCC group(P>0.05).The expression of Shh and p-SATA3 in cervical cancer tissues was related to the clinical stage of tumor,the degree of tumor differentiation and lymph node metastasis(P<0.05),and not related to tumor size and age(P>0.05).The positive expression rate of p-STAT3 was closely related to Shh expression(r=0.448,P<0.05).Conclusion The signaling pathway of Shh and STAT3 signaling pathway are related to the occurrence and progression of cervical tumors,and the activation of the two signaling pathways is synergistic.
