康莱特注射液(薏苡仁)对人肝癌细胞株SMMC7721自噬和凋亡的影响

Effects of Kanglaite injection(Coix seed)on autophagy and apoptosis of human liver cancer cell line SMMC7721

目的探讨康莱特注射液对人肝癌细胞株SMMC7721自噬和凋亡的影响。方法使用康莱特注射液处理人肝癌细胞株SMMC7721,用CCK-8法检测人肝癌细胞株SMMC7721的增殖,流式细胞仪检测凋亡率,通过Western blot检测自噬相关蛋白Beclin-1、LC3-Ⅱ等表达情况。结果与对照组比较,随着药物作用时间的延长,康莱特注射液对细胞的抑制程度逐步增加,细胞株在加药后96h表现出较好的抑制率,实验组人肝癌细胞株SMMC7721凋亡显著增加,自噬相关基因Beclin-1、LC3的mRNA与蛋白表达上调。结论康莱特注射液可能通过上调Beclin-1、LC3 mRNA水平以及蛋白表达水平,促进人肝癌细胞SMMC7721自噬,抑制其增殖,并诱导肿瘤细胞凋亡,从而抑制肿瘤生长。

Objective To investigate the effect of Kanglaite injection on autophagy and apoptosis of human liver cancer cell line SMMC7721.Methods Kanglaite injection was used to treat human hepatocellular carcinoma cell line SMMC7721.CCK-8 method was applied to detect proliferation of human hepatocellular carcinoma cell line SMMC7721,and flow cytometry was applied to detect apoptosis rate of the cell line.Western blot was applied to detect the expressions of Beclin-1,LC3-Ⅱand other autophagy-related proteins.Results Comparing with the control group,the inhibition degree of Kanglaite injection increased gradually with the prolongation of drug action time.The cell line showed a good inhibition rate 96 h after drug addition.The apoptosis of human hepatoma cell line SMMC7721 in the experimental group was significantly increased.The mRNA and protein expression of autophagy related genes beclin-1 and LC3 were up-regulated.Conclusion Kanglaite injection may promote autophagy of human hepatocellular carcinoma cell line SMMC7721 by up-regulating Beclin-1,LC3 mRNA levels and protein expression levels,inhibit its proliferation,and induce tumor cell apoptosis,thereby inhibiting tumor growth.
