摘要
目的:建立藏药三果汤中没食子基儿茶素没食子酸酯(GCG)和表没食子基儿茶素没食子酸酯(EGCG)的HPLC含量测定方法。方法:Agilent Zorbax C18键合硅胶柱(150 mm×4.6 mm,5μm);乙腈-0.1%磷酸梯度洗脱;流速:1.0 mL·min^(-1);检测波长:280 nm;柱温:25℃;进样量:10μL。结果:EGCG和GCG在浓度0.0231~0.2310 mg·mL^(-1)、0.0084~0.0840 mg·mL^(-1)范围内呈良好的线性关系,相关系数均大于0.9990;加样回收率分别为100.09%和99.52%,RSD值分别为0.95%和1.03%(n=6)。结论:建立HPLC法同时检测藏药三果汤中EGCG和GCG的含量,该方法简便、准确、重复性好,结果可靠,可为藏药三果汤中相关成分的质控提供参考。
Objective A high performance liquid chromatography(HPLC)method was established for the determination of epigallocatechin gallate(EGCG)and gallocatechin gallate(GCG)in Tibetan medicine Triphala Decoction.Methods The chromatographic column is an Agilent Zorbax C18 bonded silica column(150 mm×4.6 mm,5μm),the mobile phase is acetonitrile-0.1%phosphoric acid gradient elution,the flow rate is 1.0 mL·min^(-1),the detection wavelength is 280 nm,the column temperature is 25℃,and the injection volume is 10μL.Results EGCG and GCG showed a good linear relationship in the concentration range of 0.0231~0.2310 mL·min^(-1) and 0.0084~0.0840 mL·min^(-1),and the correlation coefficients were both greater than 0.9990;the sample recovery rates were 100.09%and 99.52%,respectively,and the RSD value They are 0.95%and 1.03%respectively(n=6).Conciusion An HPLC method was established to simultaneously detect the contents of EGCG and GCG in Tibetan medicine Sanguo Decoction.This method is simple,accurate,reproducible,and reliable.It can provide a reference for the quality control of related components in Tibetan medicine Triphala Decoction.
