摘要
枣咔实蝇Carpomya vesuviana Costa是枣树果实的一种重要检疫性害虫,其寄主枣树在我国种植广泛,该虫一旦传入和扩散,将对我国新疆红枣产业造成严重危害。本研究以枣咔实蝇蛹和成虫为对象,通过提取枣咔实蝇蛹和成虫样品的基因组DNA,借助生物信息学分析软件挖掘枣咔实蝇ITS2、16S rRNA、CYT-B-D、CYT-B-C、COI-5'、COI-3'、COII等分子鉴定靶点序列并设计扩增引物,以建立枣咔实蝇的快速分子鉴定技术。PCR扩增、测序和序列相似性结果显示,从枣咔实蝇蛹和成虫中获得的16条序列(含14条新序列),分析表明其中CYT-B、COI、COII可作为枣咔实蝇分子鉴定序列。该技术可应用于枣咔实蝇的识别鉴定、实时监测、早期防治等项工作。
Carpomya vesuviana is an important quarantine pest harming jujube trees planted widely in China.Once the pest is introduced and spread,the jujube industry in Xinjiang would be harmed seriously.The pupae and adults of C.vesuviana as the subjects were studied in present study.The genomic DNA of subjects were extracted,the molecular identification target sequences of C.vesuviana were searched including ITS2,16S rRNA,CYT-B-D,CYT-B-C,COI-5',COI-3',COII&c.,and then primers were designed for establishment of rapid molecular identification technology of C.vesuviana.PCR amplifying,sequencing and sequence similarity revealed that the 16 sequences get from pupae and adults of C.vesuviana(including 14 new sequences)can all be used as molecular identification sequence of C.vesuviana.This technique can be used in identification,real-time monitoring and early alarming or control strategies of C.vesuviana.
