摘要
目的研究甲状腺癌细胞株中miR-142-3p的表达水平,探讨miR-142-3p不同表达水平对甲状腺癌细胞增殖能力的影响。方法采用RT-PCR检测人甲状腺癌细胞K1、TPC-1、BCPAP和人甲状腺细胞Nthy-ori 3-1中miR-142-3p的表达水平。观察K1细胞分别转染miR-142-3p mimics,miR-142-3p inhibitor后对细胞增殖、克隆形成能力和细胞周期的影响。结果甲状腺癌细胞K1、TPC-1、BCPAP中的miR-142-3p相对表达量均低于甲状腺细胞Nthy-ori 3-1,差异均有统计学意义(P<0.05)。3种甲状腺癌细胞中K1细胞的miR-142-3p表达量最低。甲状腺癌细胞K1转染后120 h,与NC组相比,miR-142-3p mimics组细胞OD值降低,miR-142-3p inhibitor组细胞OD值增加,差异均有统计学意义(P<0.05)。甲状腺癌细胞K1转染后,与NC组相比,miR-142-3p mimics组细胞克隆形成减少,G0/G1期细胞百分比增加,Cyclin D1蛋白表达量下降;miR-142-3p inhibitor组细胞克隆形成增多,G0/G1期细胞百分比降低,Cyclin D1蛋白表达量升高;差异均有统计学意义(P<0.05)。结论MiR-142-3p在甲状腺癌细胞中呈低表达,高表达miR-142-3p可有效抑制甲状腺癌细胞的增殖能力,通过抑制Cyclin D1表达将细胞阻滞在G0/G1期。
Objective To explore the expression level of miR-142-3p in thyroid cancer cells(TCC),and to explore the effect of different expression levels of miR-142-3p on the proliferation of thyroid cancer cells.Methods RT-PCR was used to detect the expression levels of miR-142-3p in human thyroid cancer cells K1,TPC-1,BCPAP,and human thyroid cells Nthy-ori 3-1.The effects of K1 cells transfected with miR-142-3p mimics and miR-142-3p inhibitor on cell proliferation,colony formation,and cell cycle were observed.Results The relative expression of miR-142-3p in K1,TPC-1,and BCPAP were lower than those in Nthy-ori 3-1,and the difference was statistically significant(P<0.05).The expression of miR-142-3p in K1 was the lowest among the three thyroid carcinoma cells.Compared with the NC group,the OD value of the miR-142-3p mimics group was decreased and that of the miR-142-3p inhibitor group was increased at 120 h after transfection(P<0.05).Compared with the NC group,cell colony formation and Cyclin D1 protein expression were decreased and the percentage of G0/G1 phase cells was increased in the miR-142-3p mimics group after K1 transfection.Compared with the NC group,the miR-142-3p inhibitor group showed increased cell colony formation and Cyclin D1 protein expression,decreased percentage of G0/G1 phase cells,and the differences were statistically significant(P<0.05).Conclusions MiR-142-3p was low expressed in thyroid cancer cells.High expression of miR-142-3p can effectively inhibit the proliferation of thyroid cancer cells,and arrest the cells in G0/G1 phase by inhibiting the expression of Cyclin D1.
