摘要
目的:建立大环双内酯雷公藤生物碱高分辨质谱数据库(HR-MS-database),并应用于不同种属雷公藤生物碱的快速鉴定分析。方法:采用超高效液相色谱-四极杆-飞行时间质谱联用(UPLC/Q-TOFMS)技术手段,借助SCIEX公司Library View数据库平台,建立大环双内酯雷公藤生物碱高分辨质谱-数据库(HR-MS-database),并对不同种属间雷公藤生物碱进行差异分析。结果:建立大环双内酯结构高分辨质谱数据库,包含雷公藤生物碱65种;对28个产地不同种属间雷公藤生物碱样品测定结果显示,雷公藤属植物间生物碱成分差异明显,其中包含共有成分35种,东北雷公藤显现出与昆明山海棠和雷公藤存在明显差异。结论:本研究获得了一种大环双内酯雷公藤生物碱的快速识别方法,鉴别出不同种属间雷公藤生物碱的较大差异,为雷公藤属植物的药效物质基础与质量标准研究提供参考。
Objective:To establish the HR-MS-database of macrocyclic dilactone skeleton alkaloids from Tripterygium,and apply it in the fast identification and analysis of the alkaloids in Tripterygium form three Tripterygium species.Methods:Ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC/Q-TOF-MS)and LibraryView of SCIEX were used to construct the HR-MS-database of macrocyclic dilactone skeleton alkaloids in Tripterygium.The further research was performed for differences of the alkaloids in samples from three species of Tripterygium genus.Results:The HR-MS-database has been constructed,including 65 kinds of alkaloids in Tripterygium.The determination results of multiple samples from different habitats showed that the chemical composition of alkaloids in Tripterygium were significantly different,which contained 35 kinds of ingredients.And Tripterygium regelii Sprague et Takeda varied significantly from Tripterygium hypoglaucum(Levl.)Hutch and Tripterygium wilfordii Hook.f.Conclusion:The study provides an effective method for fast identification of alkaloids in Tripterygium with the macrocyclic dilactone skeleton.It is indicated that there are significant difference in alkaloids between different Tripterygium species.And it also provides reference for investigation on quality evaluation and substance basis of Tripterygium.
