退行性膝关节炎模型大鼠关节软骨核因子κB通路蛋白及神经肽的表达与关节功能的关系 被引量:1

Relation of joint function with expression of NF-κB pathway protein and neuropeptide in articular cartilage of rats model of degenerative knee arthritis

摘要 目的研究退行性膝关节炎模型大鼠关节软骨核因子κB(NF-κB)通路相关蛋白及神经肽的表达水平与关节功能的关系。方法建立退行性膝关节炎大鼠模型,并设健康对照组大鼠,分别检测2组大鼠的膝关节功能指标、关节软骨中NF-κB通路相关蛋白的相对表达以及神经肽的表达水平。对各指标进行Pearson相关性分析和多元线性回归分析。结果模型组大鼠攀爬时间和最长运动距离分别为(615.11±83.27)s、(58.68±17.23)m,均低于对照组大鼠[(1982.23±110.25)s、(198.21±29.96)m],而p-p65、p-IκBα、SP和CGRP的相对表达水平分别为0.35±0.11、0.26±0.05、62.71±3.75、248.98±47.54,均高于对照组大鼠(0.13±0.03、0.09±0.01、17.43±1.20、28.81±1.46),差异均有统计学意义(P<0.05)。Pearson相关性分析显示,攀爬时间和最长运动距离与p-p65、p-IκBα、SP和CGRP的表达水平呈正相关(P<0.05)。多元线性回归分析显示,p-p65(β=0.093,P<0.001)、p-IκBα(β=0.026,P<0.001)、SP(β=0.106,P<0.001)和CGRP(β=0.123,P<0.001)是攀爬时间的独立影响因素;p-p65(β=0.514,P<0.001)、p-IκBα(β=0.023,P<0.001)、SP(β=0.612,P<0.001)和CGRP(β=0.135,P<0.001)是最长运动距离的独立影响因素。结论p-p65、p-IκBα、SP和CGRP在退行性膝关节炎模型大鼠的关节软骨中呈现高表达,且与大鼠的攀爬时间和最长运动距离密切相关;p-p65、p-IκBα、SP和CGRP均为攀爬时间和最长运动距离的独立影响因素,退行性膝关节炎的关节功能与NF-κB信号通路以及神经肽的表达呈正相关。 Objective To investigate the relation of joint function with expression of nuclear factor-κB(NF-κB)pathway protein and neuropeptide in articular cartilage of rats model of degenerative knee arthritis.Methods Rats model of degenerative knee arthritis was established and healthy control group wasa set.The joint function,expression of NF-κB pathway protein and neuropeptide in articular cartilage of two groups were detected.The correlation analysis and multiple linear regression analysis were conducted.Results The climbing time and longest movement distance of rats in the model group were(615.11±83.27)s and(58.68±17.23)m,respectively,which were lower than those in the control group[(1982.23±110.25)s,(198.21±29.96)m],and the levels of p-p65,p-IκBα,SP and CGRP relative expression were 0.35±0.11,0.26±0.05,62.71±3.75,248.98±47.54,respectively,which were all higher than those of control group(0.13±0.03,0.09±0.01,17.43±1.20,28.81±1.46),the differences were statistically significant(P<0.05).The correlation analysis showed that the climbing time and maximum travel distance were positively correlated with expression of p-p65、p-IκBα、SP and CGRP(P<0.05).Multiple linear regression analysis showed that the climbing time was independently associated with p-p65(β=0.093,P<0.001),p-IκBα(β=0.026,P<0.001),SP(β=0.106,P<0.001)and CGRP(β=0.123,P<0.001);the maximum travel distance was independently associated with p-p65(β=0.514,P<0.001),p-IκBα(β=0.023,P<0.001),SP(β=0.612,P<0.001)and CGRP(β=0.135,P<0.001).Conclusion The p-p65,p-IκBα,SP and CGRP were overexpression in articular cartilage of degenerative knee arthritis,which was closely correlated with climbing time and maximum travel distance;the climbing time and maximum travel distance were independently associated with p-p65,p-IκBα,SP and CGRP,the joint function of degenerative knee arthritis was positively correlated with the expression of NF-κB pathway protein and neuropeptide.
