摘要
目的探讨白细胞介素(IL)-13通过下调紧密连接蛋白-18(Claudin-18)蛋白水平对过敏性哮喘的相关性与作用机制。方法将C57BL/6小鼠采用随机数字表法分为对照组、IL-13抑制组、模型组,每组12只。采用卵清蛋白(OVA)建立过敏性哮喘模型,21 d后取小鼠肺组织进行相关实验。采用呼吸曲线Penh值进行哮喘模型小鼠气道高反应性检测(AHR);采用实时荧光定量聚合链反应(RT-qPCR)测定Claudin-18蛋白水平,采用ELISA双抗夹心法测定IL-13与IgE水平,比较组间差异及相关性。结果模型组的AHR显著高于对照组与IL-13抑制组(P<0.05);模型组的IgE水平显著高于对照组与IL-13抑制组(P<0.05);对照组与抑制组的Claudin-18蛋白水平显著高于模型组(P<0.05);模型组的IL-13水平显著高于对照组与IL-13抑制组(P<0.05);相关性分析结果显示,Claudin-18与IL-13、IgE、AHR呈显著的负相关关系;进一步将数据标准化后以Claudin-18为因变量、IL-13为自变量建立线性回归分析可得Y=-0.003X+4.021。结论IL-13与Claudin-18存在显著的负相关线性关系,并能够通过下调Claudin-18蛋白水平而促进过敏性哮喘气道高反应性。
Objective To investigate the correlation and mechanism of IL-13 on allergic asthma by down-regulating Claudin-18 protein level.Methods The C57BL/6 mice were randomly divided into the control group,IL-13 inhibition group and model group by adopting the random number table method,12 cases in each group.Ovalbumin(OVA)was used to establish the allergic asthma model,and the mouse lung tissue was taken for conducting the relevant experiments after 21 d.The airway hyperreactivity(AHR)test was conducted by adopting the respiratory curve Penh value,the relative content of Claudin-18 protein was determined by RT qPCR,and the levels of IL-13 and IgE were determined by ELISA double antibody sandwich method.The difference and correlation between the two groups were compared.Results AHR of the model group was significantly higher than that of the control group and IL-13 inhibition group(P<0.05);the IgE level of the model group was significantly higher than that of the control group and IL-13 inhibition group(P<0.05);the level of Claudin-18 of the control group and inhibition group was significantly higher than that of the model group(P<0.05);IL-13 level of model group was significantly higher than that of the control group and IL-13 inhibition group(P<0.05);the correlation analysis results showed that Claudin-18 had significantly negative correlation with IL-13,IgE and AHR;after further standardizing the data,Claudin-18 served as the dependent variable and IL13 as the independent variable,the established linear regression analysis could obtained Y=-0.003X+4.021.Conclusion There is a significantly negative linear correlation betweenIL-13 and Claudin-18,and IL-13 can promote airway hyperresponsiveness in allergic asthma by down-regulating the Claudin-18 protein level.
