肝细胞生长因子对大鼠大脑中动脉梗塞后凋亡变化的实验分析 被引量:1

摘要 目的探讨大鼠大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)模型中肝细胞生长因子(hepatocyte growth factor,HGF)对凋亡影响的作用。方法62只SD大鼠随机分为Sham组、MCAO组及HGF组,MCAO组及HGF组均采取线栓法建立MCAO模型。各组大鼠分别采用行为学评分、TTC染色、免疫荧光、TUNEL染色、Western blot等方法检测大鼠行为学功能、梗塞面积及凋亡病理等变化。结果大鼠MCAO模型建立后HGF的受体c-Met表达升高;免疫荧光共染显示,c-Met在大鼠脑组织神经元中表达,HGF给药组梗塞面积较MCAO组减小且神经功能评分升高,TUNEL凋亡细胞在HGF给药组明显少于MCAO组。结论HGF在大鼠脑梗塞后能经过c-Met受体产生抗凋亡作用,其可能是治疗脑梗塞机制之一。 Purpose To study the changes of hepatocyte growth factor(HGF)on apoptosis in middle cerebral artery occlusion(MCAO)in rats.Methods Sixty-two SD rats were divided into Sham group,MCAO group and HGF group.MACO model was established in rats by using thread thrombosis in both MCAO group and HGF group.Behavior score,TTC staining,immunofluorescence,TUNEL staining and Western blot were to test the behavior function,infarction and apoptosis of rats in each group.Results The expression of HGF receptor c-Met enhanced after the establishement of MCAO in rats.Immunofluorescence co-staining showed that c-Met was expressed in neurons in rat brain.Compared to MCAO group,the neurobehavioral score increased,but infarction area and TUNEL positive cell decreased in HGF group.Conclusion HGF can produce anti-apoptotic effect through c-Met on MCAO in rats,which it may be a therapeutic way for ischemic stroke.
