摘要
目的:通过观察生长刺激表达基因2蛋白(ST2)在新生小鼠缺氧缺血性脑损伤(HIBD)脑内的表达及定位,初步探究IL-33/ST2轴信号通路在HIBD中的作用。方法:将20只7 d龄C57BL/6小鼠随机分配在假手术组(Sham)与HIBD模型组(HIBD),每组各10只,选用改良的Rice-Vannucci法制作HIBD动物模型,2,3,5-三苯基氯化四氮唑(TTC)染色评定脑梗死状况,采用免疫荧光组织化学染色法观察ST2阳性细胞在新生鼠HIBD模型脑内的表达情况。结果:成功建立新生小鼠HIBD模型,与Sham组相比,HIBD小鼠脑内ST2表达增加,ST2在神经元、星形胶质细胞和小胶质细胞均有表达。结论:HIBD小鼠脑内ST2表达增加,ST2可能参与缺氧缺血性脑损伤的病理进程。
Objective: Preliminarily explore the role of IL-33/ST2 axis signaling pathway that plays in hypoxic-ischemic brain damage( HIBD) by observing the expression and cell location of growth stimulation expressed gene 2( ST2) in the brain of neonatal mice with hypoxic ischemic brain damage. Methods: Twenty seven-day-old C57 BL/6 mice aged 7 d were randomly assigned to the Sham group and the HIBD group( n = 10). The HIBD model was established by the modified Rice-Vannucci method. 2,3,5-triphenyltetrazolium chloride( TTC) staining was used to evaluate the status of cerebral infarction,and immunofluorescence histochemical staining was used to detect the expression of ST2 positive cells in the HIBD model of new born mice. Results: HIBD model was well established. The expression of ST2 in HIBD neonatal mice brain that underwent HI was increased,compared with Sham group. ST2 was expressed on the cells of neurons,astrocytes,and microglia. Conclusion: The expression of ST2 is elevated in the brain of HIBD neonatal mice. ST2 is likely to be involved in the process of hypoxic ischemia brain injury in the neonatal mice.
