摘要
目的探讨经皮穴位电刺激(transcutaneous electrical acupoint stimulation,TAES)不同时间对利多卡因所致心脏毒性的保护机制。方法健康雄性大鼠50只随机均分为5组空白组、模型组、TEASⅠ组、TEASⅡ组、TEASⅢ组。选用双侧“内关”“合谷”“足三里”“三阴交”,用经皮电刺激仪持续做经皮穴位电刺激,TEASⅠ组20 min、TEASⅡ组40 min,TEASⅢ组60 min。电刺激后,各组经股静脉匀速泵注利多卡因,空白组泵注生理盐水。持续监测MAP和HR变化,检测泵注利多卡因6 min后心肌组织乳酸脱氢酶(LDH)和肌酸激酶同工酶(CK-MB)活性、总超氧化物歧化酶(T-SOD)活力、丙二醛(MDA)含量、一氧化氮(NO)含量和一氧化氮合酶(NOS)活性、Ca^(2+)含量的变化。结果注射利多卡因后大鼠心肌组织中LDH、CK-MB活性和MDA含量显著升高,Ca^(2+)含量、T-SOD活力、NO含量和NOS活性显著降低。TEAS可以显著降低LDH、CK-MB活性和MDA含量,升高T-SOD活力、NO含量和NOS活性,并且随着TEAS时间的增加,Ca^(2+)荧光强度也逐渐减弱。结论TEAS可以减轻利多卡因对心脏的毒性,而且长时间(60 min)刺激的效果更明显。
Objective To investigate the protective mechanism of different times of transcutaneous acupoint electrical stimulation(TAES)on cardiotoxicity induced by lidocaine.Methods 50 healthy male rats were randomized into five groups:blank group,model group,TEAS I group,TEAS II group and TEAS III group.We selected bilateral acupoints of"Neiguan","Hegu","Zusanli"and"Sanyinjiao",and continuously performed TEAS with transcutaneous acupoint electrical stimulation in TEAS I group(20 min),TEAS II group(40 min),and TEAS III group(60 min).After electrical stimulation,each group was pumped with lidocaine through the femoral vein,and the blank group was pumped with saline.MAP and HR changes were continuously monitored,and then we detected myocardial tissue lactate dehydrogenase(LDH),creatine kinase isoenzyme(CK-MB)activity,total superoxide dismutase(T-SOD)activity,malondialdehyde(MDA)content,nitric oxide(NO)content,nitric oxide synthase(NOS)activity and Ca^(2+)content after 6 minutes.Results The activities of LDH,CK-MB and MDA in the myocardial tissue of rats were significantly increased after injection of lidocaine.The Ca^(2+)concentration,T-SOD activity,NO content and NOS activity were significantly decreased.TEAS significantly reduced the activity of LDH,CK-MB and MDA,and significantly increased the activity of T-SOD,NO content and NOS activity,and the fluorescence intensity of Ca^(2+)was gradually weakened with the increase of TEAS time.Conclusion TEAS can reduce the toxicity of lidocaine to the heart,and the effect of a long-term stimulation(60 min)is more obvious.
