摘要
目的检测并分析五种口腔粘接剂的细胞毒性和可浸提物。方法小鼠成纤维细胞L929在五种样品的4种浓度浸提液(100%、50%、25%、12.5%)中作用24 h后,在光学显微镜下观察细胞形态变化,MTT法测定细胞相对生存率(RGR);用气相色谱质谱联用法检测固化样品细胞培养基浸提液及固化样品中可浸提物及樟脑醌含量。结果五种样品均检测出多种可浸提物,其中样品5的可浸提物的量最少,樟脑醌含量也最少,细胞毒性最小,在25%浸提液浓度时RGR为72%;其余四种样品在12.5%浸提液浓度时RGR小于39%。结论口腔粘接剂的细胞毒性与其可浸提物的量相关,优化配方、减少可浸提物的量是降低细胞毒性的有效方法。
Objective To test and analyze the cytotoxicity and their extract of five oral adhesives.Methods Mouse fibroblasts cell(L929)was treated with different extracts(100%,50%,25%,12.5%)of five samples for 24 hours.The morphology of the cells was observed by microscope.Cell relative growth rate(RGR)was tested by MTT assay.The extract and camphorquinone content in the solidified sample cell medium extract and the solidified sample were detected by gas chromatography(GC/MS).Results Several kinds of extract were detected in all of five adhesives.The self-etching adhesive showed little cytotoxicity(the RGR was 72%)at 25%extract concentration.The other four adhesives showed obvious cytotoxicity at 12.5%extract concentration.Conclusions Adhesive cytotoxicity is related with the extractable content.0 ptimizing formulation and reducing extract are effective ways to reduce the cytotoxicity.
