基于线粒体COⅠ基因的云南野桑蚕进化分析

Phylogenetic Analysis of Bombyx mandarina from Yunnan Province Based on Mitochondrial COⅠGene

野桑蚕(Bombyx mandarina)是家蚕(Bombyx mori)的祖先,研究各地野桑蚕的进化地位,对于野桑蚕资源的收集与保存具有重要意义。对云南地区野桑蚕的线粒体细胞色素C氧化酶亚基Ⅰ(COⅠ)基因进行克隆测序,并进行相关生物信息学分析。序列比对发现,不同地域来源的野桑蚕与家蚕COⅠ基因的序列一致性高达96%以上,其中云南地区同四川洪雅地区来源的野桑蚕与家蚕COⅠ基因的部分位点存在单碱基差异,且差异主要集中于A+T丰富区。系统进化分析发现,云南野桑蚕与山东青州野桑蚕的亲缘关系较近,而与地理位置相对最近的四川野桑蚕的亲缘关系较远。研究结果表明,利用COⅠ基因可以厘清云南野桑蚕同其他地域来源野桑蚕的亲缘关系,丰富野桑蚕种质资源。

Bombyx mandarina is the ancestor of silkworm(Bombyx mori).It is of great significance to study the evolution of Bombyx mandarina in various places for the collection and preservation of Bombyx mandarina resources.The mitochondrial cytochrome C oxidase subunitⅠgene(COⅠ)of Bombyx mandarina in Yunnan Province was cloned and sequenced,and the related bioinformatics analysis was performed.Sequence alignment showed that the sequence identity of COⅠgene was over 96%between Bombyx mandarina from different regions and Bombyx mori.There was a singlebase difference in partial loci of the COⅠgene between Bombyx mori and Bombyx mandarina from Yunnan Province and Hongya of Sichuan Province,and the difference was mainly concentrated in the A+T-rich region.Phylogenetic analysis found that Yunnan Bombyx mandarina was closely related to Bombyx mandarina from Qingzhou of Shandong Province,while it was far away from Bombyx mandarina from Sichuan Province which was geographical close.The research results showed that COⅠgenes could be used for clarifying the genetic relationship between Yunnan Bombyx mandarinas and Bombyx mandarinas from other different regions,and also could enrich the genetic diversity of Bombyx mandarina germplasm resources.
