摘要
作为分子伴侣系统之一,HSC/HSP70在细胞许多生理生化通路中起着十分重要的作用。杆状病毒的感染诱导宿主细胞HSC/HSP70的表达,但对HSC/HSP70在家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,BmNPV)生命周期中的作用的研究仍较少。利用特异性染料,本研究发现BmNPV表达的多种蛋白质在细胞质中形成聚集体。运用槲皮苷抑制HSC/HSP70表达后,BmNPV基因组复制、多角体蛋白表达量以及子代芽生病毒和多角体产量都显著降低。这些结果表明HSC/HSP70对BmNPV的增殖起着非常重要的作用。
Host heat shock cognate/protein of the 70 kD family(HSC/HSP70)as one of the molecular chaperones play an important role in various physiological and biochemical pathways.There are many researches on the induction of HSC/HSP70 in baculovirus infected cells.However,there are few on the role of HSC/HSP70s in infection cycle of Bombyx mori nucleopolyhedrovirus(BmNPV).In this study,production of aggregates and aggresomes formed by BmNPV proteins was found in cytoplasm,which was detected with a fluorescent molecular dye.After suppression of HSC/HSP70 expression by quercetin,genomic DNA replication of BmNPV,expression level of polyhedrin,production of budded virus(BV)and polyhedra were all significantly inhibited,indicating the dependence of BmNPV reproduction on host HSC/HSP70.
